Abstract

Penelitian ini bertujuan untuk mendapatkan data kandungan senyawa metabolit sekunder dari ekstrak dan fraksi, dan hasil uji aktivitas penghambatan enzim αglukosidase daun gofasa (Vitex cofassus Reinw.) asal Bogor, Jawa Barat. Penghambatan enzim α-glukosidase diketahui berperan sebagai agen terapeutik untuk pengobatan Diabetes Melitus tipe 2. Senyawa metabolit sekunder yang terkandung dalam tanaman ini diantaranya golongan flavonoid, fenolik, alkaloid, saponin, tanin, dan terpenoid. Ekstrak metanol, fraksi n-heksana, fraksi nheksana:etil asetat, dan fraksi etil asetat daun Vitex cofassus Reinw. diuji aktivitas penghambatan enzim α-glukosidase secara in vitro menggunakan substrat pnitrofenil-α-glukopiranosa. Hasil uji aktivitas penghambatan enzim α-glukosidase menunjukkan bahwa ekstrak metanol dan fraksi etil asetat memiliki nilai persen inhibisi yang positif, akan tetapi tidak dapat menghambat enzim α-glukosidase karena persen inhibisi yang didapat kurang dari 50%. Nilai IC50 dari ekstrak metanol adalah 22.641,229 ppm, dan nilai IC50 fraksi etil asetat adalah 628,202 ppm, sehingga keduanya dikategorikan tidak memiliki aktif sebagai zat antidiabetes. The aim of this research is to get the data of secondary metabolite compound from extract and fraction, and result of inhibition activity of α-glucosidase enzyme from gofasa leaf (Vitex cofassus Reinw.) origin Bogor, West Java. The α-glucosidase enzyme inhibitors are known to act as therapeutic agents for the treatment of type 2 diabetes mellitus. Secondary metabolite compounds contained in this plant include flavonoid, phenolic, alkaloid, saponin, tannin, and terpenoid groups. Methanol extract, n-hexane fraction, n-hexane:ethyl acetate fraction, and ethyl acetate fraction of Vitex cofassus Reinw leaf. Tested the inhibitory activity of αglucosidase enzyme in vitro using p-nitrophenyl-α-glucopyranose substrate. The results of inhibitory activity test of α-glucosidase enzyme showed that methanol extract and ethyl acetate fraction had positive percent inhibitory value, but could not inhibit α-glucosidase enzyme because percent of inhibition was less than 50%. The IC50 value of methanol extract was 22,641,229 ppm, and the IC50 value of ethyl acetate fraction was 628,202 ppm, so both were categorized as not having active as an antidiabetic agent.
