Abstract

Diabetes Melitus merupakan penyakit metabolik yang menjadi salah satu penyakit serius di Indonesia, terutama diabetes melitus tipe 2. Pencegahan DM tipe 2 dapat dilakukan dengan menghambat enzim α-glukosidase sehingga absorpsi glukosa tertunda dan kadar glukosa darah dapat menurun. Penelitian ini bertujuan untuk mengetahui aktivitas inhibisi enzim α-glukosidase dari ekstrak metanol, fraksi n-heksana, fraksi n-heksana : etil asetat, dan fraksi etil asetat daun Vitex trifolia L asal Yogyakarta. Uji aktivitas inhibisi enzim α-glukosidase dilakukan secara in vitro dengan menggunakan substrat p-nitrofenil-αglukopiranosida dan mengukur absorbansi p-nitrofenol sebagai produk reaksi enzimatis pada panjang gelombang 410 nm. Hasil penelitian menunjukkan bahwa fraksi etil asetat mengandung senyawa golongan flavonoid dan saponin. Nilai IC50 dari fraksi etil asetat yang dihasilkan adalah 691,8 ppm dan dikategorikan tidak aktif sebagai antidiabetes ketika dibandingkan dengan glukobay (sebagai kontrol) yang menghasilkan nilai IC50 sebesar 0,3259 ppm. Diabetes Mellitus is a metabolic disorder that has become a serious desease in Indonesia, especially type 2 DM. Prevention of Type 2 diabetes could be done by inhibiting α-glucosidase enzymes so that resulting delayed of glucose absorption and glucose blood leveles were decrease. The aim of this research is to know the activity of inhibition of α-glucosidase enzyme from methanol extract, nhexane fraction, n-hexane fraction: ethyl acetate, and ethyl acetate fraction of Vitex trifolia L leaves from Yogyakarta. The α-glucosidase enzyme inhibition test was performed in vitro using a p-nitrophenyl-α-glucopyranoside substrate and measured the absorbance of p-nitrophenol as a product of enzymatic reaction at 410 nm wavelength. The results showed that the fraction of ethyl acetate contains compound of flavonoid and saponin group. The IC50 value of the resulting ethyl acetate fraction was 691.8 ppm and was categorized as inactive as antidiabetic when compared with glucobay (as control) which resulted in an IC50 value of 0.3259 ppm.
