Abstract

Kedelai merupakan komoditas tanaman pangan yang saat ini mengalami penurunan produktivitas akibat keterbatasan lahan tanam, keterbatasan stok benih, dan penggunaan benih bermutu rendah. Perkecambahan dan pertumbuhan tanaman kedelai secara ex vitro maupun in vitro membutuhkan benih bermutu tinggi, media pertumbuhan kaya nutrisi, dan mengandung zat pengatur tumbuh. Penelitian ini bertujuan mengetahui pengaruh varietas dan ekstrak kecambah kacang hijau terhadap perkecambahan dan pertumbuhan kedelai varietas Anjasmoro dan Dega 1 secara ex vitro dan in vitro. Percobaan dilakukan pada bulan Mei - Desember 2020 di Laboratorium Kultur Jaringan Tumbuhan, FMIPA Universitas Negeri Jakarta dan di halaman rumah. Penelitian terdiri dari: (1) perkecambahan benih Anjasmoro dan Dega 1 secara ex vitro (substrat tanah dan kertas) dan (2) perkecambahan benih Anjasmoro dan Dega 1 secara in vitro dengan kultur embrio. Daya berkecambah benih Anjasmoro mencapai 92% dan Dega 1 sebesar 70%. Pada percobaan ex vitro di substrat tanah didapatkan Anjasmoro dengan penambahan ekstrak kecambah kacang hijau 450 gr/L menghasilkan pertumbuhan terbaik pada tinggi tanaman, jumlah cabang, jumlah buku batang utama, jumlah daun, dan jumlah bunga. Pertumbuhan jumlah cabang, jumlah buku, jumlah daun, dan jumlah bunga terbaik pada kedelai yang dikecambahkan di substrat kertas ditemukan pada Dega 1 dengan penambahan ekstrak kecambah kacang hijau 450 gr/L. Pertumbuhan kedelai in vitro hasil kultur embrio menunjukkan bahwa Dega 1 dalam media tanam MS 0 unggul pada parameter tinggi tanaman, jumlah akar, jumlah cabang, dan jumlah daun. Soybean is a food crop commodity which is currently experiencing a decline in productivity due to limited land for planting, limited seed stocks, and the use of low quality seeds. Germination and growth of soybean plants both ex vitro and in vitro require high quality seeds, nutrient-rich growth media, and contain plant growth regulator. This study aims to determine the effect of varieties and extracts of green bean sprouts on the germination and growth of soybean varieties Anjasmoro and Dega 1 ex vitro and in vitro. The experiment was carried out in May - December 2020 at the Plant Tissue Culture Laboratory, Faculty of Mathematic and Natural Science, State University of Jakarta and on the home yard. The research consisted of: (1) germination of Anjasmoro and Dega 1 seeds by ex vitro (soil and paper substrate) and (2) germination of seeds of Anjasmoro and Dega 1 in vitro using embryo culture technique. Seed germination of Anjasmoro reached 92% and Dega 1 was 70%. In experiments ex vitro on soil substrates, it was found that the Anjasmoro variety with the addition of 450 gr/L green bean sprout extract produced the best growth for plant height, number of branches, number of main stem nodes, number of leaves, and number of flowers. The best growth in the number of branches, number of nodes, number of leaves, and number of flowers on soybeans germinated on paper substrate was found in Dega 1 variety with the addition of 450 gr/L green bean sprout extract. The growth of soybean in vitro from embryo culture showed that the Dega 1 in MS 0 growing medium was superior to the parameters of plant height, number of roots, number of branches, and number of leaves
