Abstract

Aluminium (Al) merupakan logam non-esensial dan bersifat toksik bagi tubuh. Aluminium memiliki waktu paruh 7 tahun, sehingga waktu yang diperlukan untuk keluar dari tubuh sangat lama. Disamping waktu paruh yang lama, aluminium memiliki sifat afinitas yang lebih besar daripada besi, sehingga Al mampu masuk kedalam tubuh dan bersaing dengan besi untuk berikatan dengan transferrin. Logam ini akan terakumulasi di tubuh menimbulkan efek toksisitas pada organ manusia, salah satunya di ginjal. Aluminium yang terakumulasi di ginjal akan menyebabkan nefrotoksisitas dan stres oksidatif melalui produksi Reactive Oxygen Species (ROS). Salah satu senyawa yang dapat menghambat stres oksidatif adalah resveratrol. Resveratrol hasil isolasi anggur telah terbukti mempunyai aktivitas nefroprotektif dan dapat menghambat stres oksidatif. Di Indonesia, resveratrol berhasil di isolasi dari tempe, tetapi harus diuji terlebih dahulu secara in vivo aktivitas nefroprotektifnya. Metode yang digunakan adalah Rancangan Acak Lengkap (RAL), dosis AlCl₃ yang digunakan 200 mg/kgBB dan dosis resveratrol tempe yang digunakan 5 dan 10 mg/kgBB. Data yang diperoleh adalah gambaran jaringan histologis ginjal meliputi atrofi glomerulus dan degenerasi tubulus, kadar MDA serta aktivititas katalase. Hasil yang diperoleh yaitu resveratrol tempe dosis 5 dan 10 mg/kgBB mempunyai efek nefroprotektif pada ginjal mencit umur dua bulan yang diberikan aluminium klorida dosis 200 mg/kgBB. Aluminum (Al) is a non-essential metal and is toxic to the body. Aluminum has a halflife time of 7 years, which takes a very long time to leave the body. Besides the long half-life time, aluminum has a greater affinity than iron, causing Al to be able to enter the body and compete with iron for binding to transferrin. This metal will accumulate in the body causing toxic effects on human organs, one of which is in the kidney. Aluminum that accumulates in the kidney will cause nephrotoxicity and oxidative stress through the production of Reactive Oxygen Species (ROS). One of the compounds that can inhibit oxidative stress is resveratrol. Resveratrol isolated from grapes has been shown to have nephroprotective activity and can inhibit oxidative stress. In Indonesia, resveratrol has been isolated from tempe, but it must first be tested through in vivo for its nephroprotective activity. The method used was a completely randomized design (CRD), the dose of AlCl₃ used was 200 mg/kgBW and the dose of resveratrol tempe used were 5 and 10 mg/kgBW. The data obtained were histological features of the kidney including glomerular atrophy and tubular degeneration, MDA levels and catalase activity. The results obtained were resveratrol tempe at doses of 5 and 10 mg/kgBW had a nephroprotective effect on the kidney of two-month-old mice given aluminum chloride at a dose of 200 mg/kgBW.
