Abstract

7-hidroksi-4-metilkumarin merupakan senyawa turunan kumarin yang sangat diminati dalam sintesis organik karena aktivitas biologisnya yang signifikan. Pada penelitian ini dilakukan optimasi kondisi reaksi sintesis 7-hidroksi-4-metilkumarin menggunakan metode refluks dengan mereaksikan resorsinol dan etil asetoasetat dalam pelarut etanol selama 3 jam dan 6 jam yang dimonitoring dengan kromatografi lapis tipis. Optimasi dilakukan dengan berbagai jumlah katalis yang bervariasi. Pemurnian crude product dilakukan dengan rekristalisasi. Berdasarkan hasil penelitian, diperoleh yield 7-hidroksi-4-metilkumarin berturutturut sebesar 8%, 18%, 28% dan 47%. Produk yang dihasilkan pada sintesis ini diuji titik lelehnya dan ditentukan strukturnya secara spektroskopi UV dan 1H-NMR. 7-hydroxy-4-methylcoumarin is a coumarin derivative which is great interest in organic synthesis due to its significant biological activity. In this study, optimization of the reaction conditions for the synthesis of 7-hydroxy-4-methylcoumarin was carried out using the reflux method by reacting resorcinol and ethyl acetoacetate in ethanol for 3 hours and 6 hours monitored by thin layer chromatography. Optimization is carried out with various amounts of varying catalysts. Purification of crude product is done by recrystallization. Based on the results of the study, the yield of 7-hydroxy-4-methylcoumarin was 8%, 18%, 28% and 47%, respectively. The product produced in this synthesis was tested for its melting point and determined spectroscopic structure which includes UV and 1H-NMR.
