Abstract

Sapi Bali merupakan sapi asli Indonesia yang didomestikasi dari banteng liar dan merupakan sumber produksi daging sapi terbesar di Indonesia. Penelitian ini dilakukan untuk mengetahui perbedaan sekuens gen MT-ND1 dan mengetahui hubungan antara sapi Bali polled dan sapi Bali bertanduk berdasarkan gen MT-ND1. Hasil penelitian berupa sekuens MT-ND1 522 bp menunjukkan adanya variasi basa nukleotida pada sapi Bali polled berupa 36 titik transisi dan 3 titik transversi. Jarak genetik antara sapi Bali polled dengan sapi Bali bertanduk adalah 0,00 dan jarak genetik antara sapi Bali polled dengan Bos javanicus adalah 0,08. Analisis pohon filogenetik menunjukkan bahwa sapi Bali polled berada dalam clade yang sama dengan sapi Bali bertanduk (monophyly). Sedangkan Bos javanicus berada pada cluster yang berbeda dan berkerabat dekat dengan Bos taurus. Kesimpulan dari penelitian ini adalah fragmen MT-ND1 522 bp pada penelitian ini tidak dapat menjadi penanda genetik untuk membedakan sapi Bali polled dengan sapi Bali bertanduk dan berdasarkan fragmen MT-ND1 sapi Bali polled memiliki hubungan yang sangat erat dengan sapi Bali bertanduk atau memiliki keturunan maternal yang sama. ***** Bali cattle are native Indonesian cattle domesticated from wild banteng and are the largest source of beef production in Indonesia. This study was conducted to know the difference in the MT-ND1 gene sequences and knowing the relationship between polled Bali cattle and horned Bali cattle based on the MT-ND1 gene. The results of this study in the form of a 522 bp MT-ND1 sequence showed variations in nucleotide bases in polled Bali cattle in the form of 36 points of transition and 3 points of transversion. The genetic distance between polled Bali cattle and horned Bali cattle was 0.00 and the genetic distance between polled Bali cattle and Bos javanicus was 0.08. The phylogenetic tree analysis showed that the polled Bali cattle were in the same clade as the horned Bali cattle (monophyly). Meanwhile, Bos javanicus is in a different cluster and is closely related to Bos taurus. The conclusion of this study is the 522 bp MT-ND1 fragment in this study cannot be a genetic marker to distinguish polled Bali cattle from horned Bali cattle and based on the MT-ND1 fragment, Bali polled cattle have a very close relationship with horned Bali cattle or have the same maternal ancestry.
