Abstract

Tanaman berpotensi untuk dikembangkan sebagai obat tradisional. Beberapa obat tradisional digunakan masyarakat karena kandungan flavonoid dan fenol didalamnya serta aman digunakan. Temu putih (Curcuma zedoaria), mempunyai kandungan utama senyawa-senyawa kurkuminoid. Sedangkan, kandungan utama dalam kulit buah manggis (Garcinia mangostana L.) adalah xanton. Penelitian ini bertujuan untuk menganalisis kandungan senyawa flavonoid, fenol, dan sifat toksisitas dari kombinasi ekstrak temu putih (Curcuma zedoaria) dan kulit manggis (Garcinia mangostana L.) menggunakan metode Brine Shrimp Lethality Test (BSLT) yang dinyatakan dalam nilai LC50. Metode yang digunakan adalah eksperimental dengan rancangan acak lengkap. Variabel bebas penelitian berupa variasi kombinasi ekstrak temu putih dan kulit manggis (1:2, 1:1 dan 2:1), sedangkan variabel terikat penelitian meliputi kandungan kualitatif flavonoid dan fenol, kadar flavonoid dan fenol total, serta nilai LC50. Uji toksisitas dilakukan dengan desain rancang acak lengkap dengan variasi perbandingan kombinasi ekstrak dan konsentrasi (20, 50, 80, 110 dan 110 ppm). Analisis data dilakukan kualitatif secara deskriptif dan data kadar fenol, flavonoid serta toksisitas dianalisis dengan ANOVA. Hasil penetapan kadar flavonoid pada ekstrak temu putih dan manggis memiliki kandungan flavonoid dan fenol dengan kadar flavonoid 93,09 ± 0,07 mgQE/g untuk perbandingan 1:2, 86,24 ± 0,27 mgQE/g untuk perbandingan 1:1, dan 75,37 ± 0,27 mgQE/g untuk perbandingan 2:1. Kadar fenol total perbandingan 1:2, 1:1, dan 2:1 berturut-turut adalah 122,87 ± 1,64 mgGAE/g, 71,56 ± 0,57 mgGAE/g, dan 83,17 ± 0,47 mgGAE/g. Hasil uji BSLT menunjukkan bahwa ketiga variasi kombinasi ekstrak bersifat toksik dengan nilai LC50 111,76 ppm 135,58 ppm dan 81,78 ppm. ***** Plants have the potential to be developed as traditional medicine. Some traditional medicines are used by the community because they contain flavonoids and phenols in them and are safe to use. White turmeric (Curcuma zedoaria), has the main content of curcuminoid compounds. Meanwhile, the main content in the mangosteen rind (Garcinia mangostana L.) is xanthones. This study aims to analyze the content of flavonoid compounds, phenols, and toxicity of the extracts of the combination of white turmeric (Curcuma zedoaria) and mangosteen rind (Garcinia mangostana L.) using the Brine Shrimp Lethality Test (BSLT) method which is expressed in the LC50 value. The method used is experimental with a completely randomized design. The independent variables of the study were variations in the combination of extracts of white turmeric and mangosteen rind (1:2, 1:1 and 2:1), while the dependent variables of the study included qualitative content of flavonoids and phenols, levels of flavonoids and phenols, and LC50 value. Toxicity test use a completely randomized design with various combinations of extracts ratio and concentrations (20, 50, 80, 110 and 110 ppm). Data analysis was done qualitatively descriptively and data on phenol, flavonoid and toxicity levels were analyzed using ANOVA. The results of the determination of flavonoid levels in extract of white turmeric and mangosteen contained flavonoids and phenols with flavonoid levels 93,09 ± 0,07 mgQE/g for a ratio of 1:2, 86,24 ± 0,27 mgQE/g for a ratio of 1:1, and 75,37 ± 0,27 mgQE/g for a ratio of 2:1. The total phenol levels in a ratio of 1:2, 1:1, and 2:1 are 122,87 ± 1,64 mgGAE/g, 71,56 ± 0,57 mgGAE/g, and 83,17 ± 0,47 mgGAE, respectively. BSLT test result showed that the three variations of the extract combination are toxic with LC50 values of 111,76 ppm, 135,58 ppm and 81,78 ppm.
