Abstract

Kunyit (Curcuma domestica Val.), Jahe Merah (Zingiber officinale Roscoe), dan Sereh (Cymbopogon citratus (DC.) Stapf) merupakan tumbuhan yang banyak dimanfaatkan oleh masyarakat sebagai obat karena memiliki kandungan senyawa metabolit sekunder untuk pencegahan dan pengobatan penyakit. Terdapat berbagai macam bentuk pemanfaatan tumbuhan obat diantaranya dalam bentuk ekstrak, seduhan, dan rebusan. Penelitian ini bertujuan untuk mengetahui kandungan senyawa fenol, kadar total fenol, dan aktivitas antioksidan berdasarkan metode DPPH, FRAP, dan ABTS. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) dengan dua faktorial. Faktor pertama yaitu variasi jenis sediaan (ekstrak, rebusan, dan seduhan). Faktor kedua yaitu konsentrasi sampel (10, 50, 100, 150, dan 200 ppm). Asam askorbat digunakan sebagai kontrol positif dalam penelitian ini. Data dianalisis dengan ANOVA dan dilanjutkan dengan uji Duncan. Hasil skrining fitokimia menunjukkan kombinasi kunyit, jahe merah, dan sereh pada berbagai sediaan memiliki kandungan senyawa fenol. Hasil pengujian kadar total fenol yang paling optimal diperoleh pada sediaan ekstrak yaitu 21,751 ± 0.798 mgGAE/g. Hasil pengujian aktivitas antioksidan menunjukkan sediaan ekstrak merupakan sediaan yang paling optimal dalam meredam radikal bebas DPPH, FRAP, dan ABTS pada konsentrasi 200 ppm secara berturut-turut yaitu sebesar 80,84%, 54,53%, dan 79,72%. Nilai IC50 yang paling optimal sebagai antioksidan terhadap radikal DPPH, FRAP, dan ABTS secara berturut-turut yaitu 40,03 ppm, 169,63 ppm, dan 25,91 ppm. Berdasarkan hasil penelitian, kombinasi kunyit, jahe merah, dan sereh pada berbagai sediaan berpotensi sebagai sumber bahan alami dalam menghasilkan senyawa antioksidan untuk mencegah radikal bebas. Turmeric (Curcuma domestica Val.), Red Ginger (Zingiber officinale Roscoe), and Lemongrass (Cymbopogon citratus (DC.) Stapf) are plants that are widely used by the community as medicine because they contain secondary metabolites for the prevention and treatment of disease. There are various forms of utilization of medicinal plants including in the form of extracts, stew, and steepings. This study aims to determine the content of phenolic compounds, total phenol content, and antioxidant activity based on the DPPH, FRAP, and ABTS methods. This study used a completely randomized design (CRD) with two factorials. The first factor is variations in the type of preparation (extracts, stew, and steepings). The second factor is sample concentration (10, 50, 100, 150, and 200 ppm). Ascorbic acid was used as a positive control in this study. Data were analyzed by ANOVA and followed by Duncan's test. The results of phytochemical screening showed that the combination of turmeric, red ginger, and lemongrass in various preparations contained phenolic compounds. The results of the test for the most optimal total phenol content were obtained in the extract preparation, namely 21.751 ± 0.798 mgGAE/g. The results of the antioxidant activity test showed that the extract preparation was the most optimal preparation in reducing the free radicals of DPPH, FRAP, and ABTS at a concentration of 200 ppm, respectively, namely 80.84%, 54.53%, and 79.72%. DPPH free radical scavenging showed the highest results, indicating the dominance of the antioxidant activity of the extract preparation having a very strong antioxidant activity. The most optimal IC50 values as antioxidants against DPPH, FRAP, and ABTS radicals, respectively, were 40,03 ppm, 169,63 ppm, dan 25,91 ppm. Based on the research results, the combination of turmeric, red ginger, and lemongrass in various preparations has the potential as a source of natural ingredients in producing antioxidant compounds to prevent free radicals.
