Abstract

Tanaman kencur (Kaempferia galanga L.), lengkuas (Alpinia galanga L.) dan temulawak (Curcuma xanthorrhiza Roxb.) merupakan tanaman herbal yang banyak digunakan sebagai obat tradisional karena kandungan senyawa aktif didalamnya yaitu fenol dan flavonoid memiliki manfaat sebagai antioksidan. Penelitian ini bertujuan untuk mengetahui aktivitas antioksidan kombinasi ekstrak kencur, lengkuas, dan temulawak berdasarkan metode DPPH, ABTS, dan FRAP. Metode penelitian yang digunakan adalah metode deskriptif dan metode eksperimental. Metode deskriptif digunakan untuk skrining fitokimia berupa flavonoid dan fenol, serta identifikasi senyawa golongan flavonoid dari kombinasi ekstrak. Metode eksperimental digunakan untuk menganalisis aktivitas antioksidan berdasarkan metode DPPH, ABTS, dan FRAP. Desain penelitian yang digunakan adalah Rancangan Acak Lengkap (RAL) dengan dua faktorial. Faktor pertama adalah kombinasi ekstrak A (1:1:1), B (1:2:1), C (2:1:2) dan faktor kedua adalah konsentrasi (10, 50, 100, 150, 200 ppm). Data dianalisis menggunakan ANOVA dua arah dan dilanjut dengan uji Duncan. Hasil penelitian menunjukkan aktivitas antioksidan pada kombinasi ekstrak B dengan konsentrasi 200 ppm mampu meredam radikal bebas DPPH sebesar 78,21%. Aktivitas antioksidan pada kombinasi ekstrak A dengan konsentrasi 200 ppm mampu meredam radikal bebas ABTS sebesar 91,11%. Aktivitas antioksidan pada kombinasi ekstrak A dengan konsentrasi 200 ppm mampu meredam radikal bebas FRAP sebesar 52,76%. Kombinasi ekstrak kencur, lengkuas, dan temulawak berpotensi sebagai sumber antioksidan yang baik memiliki kemampuan scavenging, sedangkan pada asam askorbat sebagai sumber antioksidan yang memiliki kemampuan dalam mereduksi ion logam Fe3+ menjadi ion Fe2+. ********** Kencur (Kaempferia galanga L.), galangal (Alpinia galanga L.) and temulawak (Curcuma xanthorrhiza Roxb.) plants are herbal plants that are widely used as traditional medicine because of their active compounds phenols and flavonoids that known for their antioxidant activity. This study aims to determine the antioxidant activity of the extract combination of kencur, galangal, and curcuma based on the DPPH, ABTS, and FRAP methods. The research method used were descriptive method and experimental method. Descriptive method was used for phytochemical screening in the flavonoids and phenols, and identification of flavonoid group compounds from the combination of extracts. Experimental method was used to analyze antioxidant activity based on DPPH, ABTS, and FRAP methods. The research design used was a Completely Randomized Design (CRD) with two factorials. The first factor was a combination of extracts A (1:1:1), B (1:2:1), C (2:1:2) and the second factor was the concentration (10, 50, 100, 150, 200 ppm). Data were analyzed using two-way ANOVA and continued with Duncan's test. The results showed the antioxidant activity in the combination of extract B with 200 ppm in concentration was able to reduce DPPH free radicals by 78.21%. The antioxidant activity in the combination of extract A with 200 ppm in concentration was able to reduce ABTS free radicals by 91.11%. The antioxidant activity in the combination of extract A with a 200 ppm in concentration was able to reduce FRAP free radicals by 52.76%. The combination of kencur, galangal, and curcuma extracts has the potential to be a good source of antioxidants having scavenging abilities, while ascorbic acid is a source of antioxidants which has the ability to reduce Fe3+ metal ions to Fe2+ ions.
