Abstract

Darah adalah cairan ekstraseluler yang berperan dalam homeostasis fisiologis tubuh. Salah satu komponen utama sebagai sumber energi tubuh dalam darah adalah glukosa. Peningkatan kandungan glukosa darah dapat mengganggu sirkulasi darah dalam tubuh yang ditandai dengan peningkatan viskositas darah. Tujuan dari penelitian ini yaitu untuk mengetahui aktivitas antidiabetik ekstrak daun kaliandra terhadap hemosirkulasi pada mencit diabetes. Penelitian ini menggunakan 6 kelompok perlakuan yaitu Kontrol Sehat (mencit tanpa perlakuan apapun), Kontrol Negatif (mencit diabetes), Kontrol Positif (mencit diabetes+metformin), Perlakuan 1 (mencit diabetes+ekstrak daun kaliandra 140 mg/kg BB), Perlakuan 2 (mencit diabetes+ekstrak daun kaliandra 280 mg/kg BB), dan Perlakuan 3 (mencit diabetes+ekstrak daun kaliandra 560 mg/kg BB). Induksi aloksan dosis 210 mg/kg BB dilakukan melalui injeksi intraperitoneal pada H1 dan pengecekan kadar glukosa darah pada H4. Pemberian metformin serta ekstrak daun kaliandra pada H5 hingga H18 secara oral. Pengujian sirkulasi darah dilaksanakan pada H19 meliputi pengukuran saturasi oksigen, durasi perdarahan, kadar albumin, dan viskositas darah. Hasil penelitian menunjukkan adanya perbedaan secara signifikan pada saturasi oksigen, durasi perdarahan, dan nilai hematokrit darah, pada seluruh kelompok perlakuan dibandingkan kelompok kontrol negatif (Sig. <0,05). Kadar albumin pada kelompok perlakuan tidak memiliki perbedaan signifikan dari kelompok kontrol negatif (Sig. >0,05). Pemberian ekstrak daun kaliandra memiliki pengaruh terhadap perubahan hemosirkulasi mencit diabetes yang ditandai dengan peningkatan nilai hematokrit dan saturasi oksigen serta penurunan durasi perdarahan. ****** Blood is an extracellular fluid that plays a role in the physiological homeostasis of the body. One of the main components as the body's energy source in the blood is glucose. An increase in blood glucose content can interfere with blood circulation in the body which is characterized by an increase in blood viscosity. The purpose of this study was to determine the antidiabetic activity of calliandra leaf extract on hemocirculation in diabetic mice. This study used 6 treatment groups, namely Control (mice without any treatment), Negative Control (diabetic mice), Positive Control (diabetic mice + metformin), Treatment 1 (diabetic mice + calliandra leaf extract 140 mg/kg BW), Treatment 2 (diabetic mice + calliandra leaf extract 280 mg/kg BW), and Treatment 3 (diabetic mice + calliandra leaf extract 560 mg/kg BW). Alloxan induction at a dose of 210 mg/kg BW was carried out via intraperitoneal injection on D1 and checking blood glucose levels on D4. Administration of metformin and calliandra leaf extract at D5 to D18 orally. Blood circulation testing was carried out on D19 included measuring oxygen saturation, bleeding time, albumin levels, and blood viscosity. The results showed that there were significant differences in the value of oxygen saturation and duration of bleeding, and blood hematocrit, in all treatment groups compared to the negative control group (Sig. <0,05). Albumin levels in the treatment group did not differ significantly from the negative control group (Sig. >0,05). The administration of calliandra leaf extract had an effect on changes in the hemocirculation of diabetic mice which were characterized by an increase in the value of hematocrit and oxygen saturation and a decrease in the duration of bleeding.
