Abstract
Paparan radiasi dapat menginduksi kerusakan bahkan kematian pada sel. Sinar gamma merupakan zat radioaktif bersifat non-selektif sehingga dapat merusak sel dan jaringan yang sehat pada manusia. Resveratrol (RES) sebagai senyawa fenolik yang dapat mengurangi pembentukan radikal bebas dan berperan sebagai agen radiomitigator. RES yang diisolasi dari tempe sebagai makanan khas Indonesia terbukti dapat meningkatkan viabilitas sel. Penelitian ini bertujuan menganalisis perbedaan frekuensi mikronukleus, rasio PCE (Polychromatic Erythrocyte) terhadap NCE (Normochromatic Erythrocyte) pada sel bone marrow mencit, melaui metode In vivo Micronucleus Assay. Sebanyak 24 mencit jantan dewasa dikelompokkan secara acak menjadi enam kelompok (4 mencit/kelompok). Kelompok kontrol (K), Kelompok RES Standar (RESS), Kelompok RES tempe (REST), Kelompok Radiasi (RAD), Kelompok Radiasi+Resveratrol standar (RAD+RESS), Kelompok Radiasi+Resveratrol tempe (RAD+REST). Resveratrol dosis 10 mg/kg berat badan diberikan setelah paparan radiasi 5 Gy selama 14 hari. Mencit dibunuh pada hari ke 15 dengan anastesi menggunakan ketamin dosis 0,2 ml Hasil penelitian menunjukkan bahwa radiasi 5 Gy terbukti meningkatkan rata-rata jumlah frekuensi MN serta menurunkan indeks sitotoksik dan indeks proliferasi pada sel bone marrow mencit setelah 1 hingga 14 hari hari paska radiasi. Ini dapat disimpulkan bahwa RES Tempe bersifat sebagai agen mitigator terhadap frekuensi MN pada mencit Jantan iradiasi gamma 5 Gy. *********** Radiation exposure can induce damage and even death in cells. Gamma rays are radioactive substances that are non-selective so they can damage healthy cells and tissues in humans. Resveratrol (RES) as a phenolic compound that can reduce the formation of free radicals and acts as a radiomitigator agent. RES isolated from tempeh as a typical Indonesian food has been shown to increase cell viability. This study aims to analyze differences in micronucleus frequency, the ratio of PCE (Polychromatic Erythrocyte) to NCE (Normochromatic Erythrocyte) in mice bone marrow cells, through the In vivo Micronucleus Assay method. A total of 24 adult male mice were randomized into six groups (4 mice/group). Control group (K), Standard RES Group (RESS), Tempe RES Group (REST), Radiation Group (RAD), Radiation+Resveratrol standard group (RAD+RESS), Radiation+ Resveratrol tempeh group (RAD+REST). Resveratrol dose of 10 mg/kg body weight was given after 5 Gy radiation exposure for 14 days. The mice were killed on the 15th day under anesthesia using a dose of 0.2 ml of ketamine. The results showed that 5 Gy radiation was shown to increase the average number of MN frequencies and decrease the cytotoxic index and proliferation index in bone marrow cells of mice after 1 to 14 days after radiation. It can be concluded that RES Tempeh is a mitigating agent against MN frequency in 5 Gy gamma irradiated male mice.
