Abstract
Diabetes melitus tidak hanya dialami orang dewasa namun, anak–anak hingga remaja dapat mengalami diabetes melitus. Diabetes melitus juga dapat terjadi pada wanita usia subur yang berpengaruh pada siklus menstruasi dan fertilitas. Daun kelor dapat digunakan untuk pengobatan hiperglikemia. Penggunaan hewan model seperti mencit dalam dunia penelitian digunakan untuk tujuan tertentu diharapkan hewan tersebut mampu berperan atau menyerupai objek penelitian sesungguhnya. Penelitian ini bertujuan untuk mengetahui efek dari pemberian ekstrak daun kelor (Moringa oleifera L.) terhadap siklus estrus dan sel folikel ovarium mencit diabetes. Penelitian ini menggunakan metode eksperimental dengan desain Rancangan Acak Kelompok (RAK). Mencit betina pasca sapih strain DDY sebanyak 25 ekor dibagi menjadi 5 kelompok perlakuan yaitu Kontrol Normal, Kontrol (+), Kontrol (-), EDK 150 mg/kgBB dan EDK 200 mg/kgBB. Data dianalisis dengan Statistical Product for Social Science (SPSS) versi 25 dengan metode uji One Way ANOVA dengan taraf kepercayaan 95% dan dilanjutkan dengan Duncan. Hasil penelitian menunjukkan ekstrak daun kelor (Moringa oleifera L.) berpengaruh signifikan terhadap bobot mencit pasca sapih diabetes mellitus (Sig 0.000<0.05). Ekstrak daun kelor (Moringa oleifera L.) berpengaruh signifikan terhadap penurunan kadar glukosa darah puasa mencit pasca sapih diabetes mellitus (Sig 0.000<0.05). Ekstrak daun kelor (Moringa oleifera L.) tidak berpengaruh signifikan terhadap siklus estrus mencit pasca sapih diabetes mellitus (Sig 0.322>0.05). Ekstrak daun kelor (Moringa oleifera L.) tidak berpengaruh signifikan terhadap bobot ovarium mencit pasca sapih diabetes mellitus (Sig 0.302>0.05). Ekstrak daun kelor (Moringa oleifera L.) berpengaruh signifikan terhadap sel folikel ovarium mencit pasca sapih diabetes mellitus (Sig 0.000<0.05). Disimpulkan bahwa ekstrak daun kelor (Moringa oleifera L.) mampu menurunkan perkembangan folikel namun tidak menyebabkan perubahan siklus estrus dan bobot ovarium. Diabetes mellitus can also occur in women of childbearing age which affects the menstrual cycle and fertility. Moringa leaves can be used for the treatment of hyperglycemia. The use of model animals such as mice in the world of research is used for certain purposes, it is hoped that these animals can play a role or resemble real research objects. This study aims to determine the effect of moringa leaf extract (Moringa oleifera L.) on the estrous cycle and ovarian follicular cells of diabetic mice. This study used an experimental method with a randomized block design (RBD). 25 DDY strain post-weaning female mice were divided into 5 treatment groups namely Normal Control, Control (+), Control (-), EDK 150 mg/kgBB and EDK 200 mg/kgBB. Data were analyzed using Statistical Product for Social Science (SPSS) version 25 with the One Way ANOVA test method with a 95% confidence level and continued with Duncan. The results showed that moringa leaf extract (Moringa oleifera L.) had a significant effect on the weight of post-weaning mice with diabetes mellitus (Sig 0.000 <0.05). Moringa leaf extract (Moringa oleifera L.) has a significant effect on decreasing fasting blood glucose levels in post-weaning mice with diabetes mellitus (Sig 0.000<0.05). Moringa leaf extract (Moringa oleifera L.) had no significant effect on the estrous cycle of post-weaning mice with diabetes mellitus (Sig 0.322>0.05). Moringa leaf extract (Moringa oleifera L.) had no significant effect on the ovarian weight of post-weaning diabetes mellitus mice (Sig 0.302>0.05). Moringa leaf extract (Moringa oleifera L.) had a significant effect on the ovarian follicular cells of post-weaning mice with diabetes mellitus (Sig 0.000<0.05). It was concluded that moringa leaf extract (Moringa oleifera L.) was able to reduce follicular development but did not cause changes in estrus cycle and ovarian weight.
