Abstract
Antibiotik umum digunakan untuk mengatasi berbagai jenis penyakit infeksi. Namun demikian, penggunaan yang tidak terkendali atau tidak sesuai dosis telah menimbulkan resistensi pada mikroorganisme patogen penyebab infeksi sehingga menurunkan efektifitas antibiotik. Eksplorasi sumber antibiotik baru menjadi salah satu upaya untuk mengatasi resistensi patogen. Tanaman mengandung metabolit sekunder yang dapat berperan sebagai senyawa antibakteri. Penelitian ini bertujuan untuk mengetahui potensi antibakteri ekstrak daun simpor (Dillenia suffruticosa) asal Bangka Belitung terhadap Bacillus cereus dan Escherichia coli. Metode yang digunakan adalah deskriptif untuk analisis kualitatif dan kuantitatif tanin, fenolik dan alkaloid, dan eksperimen dengan desain Rancangan Acak Lengkap (RAL). Berbagai variasi konsentrasi esktrak daun simpor (0,05; 0,10; 0,15; 0,20; 0,25 g/mL) digunakan sebagai faktor penelitian dengan zona hambat dan bakteri bakteri Bacillus cereus dan Escherichia coli asal Kultur Koleksi Laboratorium Mikrobiologi Universitas Negeri Jakarta sebagai parameter uji. Hasil uji fitokimia ditemukan ekstrak daun simpor mengandung alkaloid 12%, fenolik 25,2 mgGAE/g dan terpenoid 5%. Hasil uji antibakteri menunjukkan bahwa konsentrasi ekstrak daun simpor 0,25 g/mL berpengaruh terhadap bakteri Bacillus cereus dengan nilai zona hambat 10,73 ± 0,85 mm. Hasil penelitian ini mengungkapkan bahwa ekstrak daun simpor dapat menghambat bakteri Bacillus cereus tetapi tidak berpengaruh terhadap bakteri Escherichia coli. *********************************************************** Antibiotics are commonly used to treat various types of infectious diseases. However, uncontrolled use or inappropriate dosage causes resistance to pathogenic microorganisms that cause infection, thereby reducing the effectiveness of antibiotics. Exploration of new sources of antibiotics is one of the efforts to overcome pathogen resistance. Plants contain secondary metabolites that can act as antibacterial compounds. This study aimed to determine the antibacterial potential of simpor leaf extract (Dillenia suffruticosa) from Bangka Belitung against Bacillus cereus and Escherichia coli. The method used is descriptive for qualitative and quantitative analysis of tannins, phenolics, and alkaloids, and experiments with Completely Randomized Design (CRD). Various variations of the concentration of simpor leaf extract (0,05; 0,10; 0,15; 0,20; 0,25 g/mL) were used as research factors with inhibition zones and bacteria Bacillus cereus and Escherichia coli from Microbiology Laboratory Collection Culture Jakarta State University as a parameter test. Phytochemical test results found simpor leaf extract contains 12% alkaloids, 25,2 mgGAE/g phenolic, and 5% terpenoids. The results of the antibacterial test showed that the concentration of simpor leaf extract 0,25 g/mL had an effect on Bacillus cereus bacteria with an inhibition zone value of 10,73 ± 0,85 mm. The results of this study revealed that simpor leaf extract could inhibit Bacillus cereus bacteria but had no effect on Escherichia coli bacteria.
