Abstract
Pisang merupakan hasil pangan pertanian yang memiliki nilai komersial tinggi dan berpeluang untuk dikembangkan. Permasalahan yang dihadapi dalam budidaya pisang adalah sulit mendapatkan bibit yang berkualitas dalam jumlah besar dalam waktu yang singkat. Tujuan penelitian ini yaitu: (1) Mengetahui dosis letal (LD50) untuk meningkatkan variasi genetik pisang Barangan setelah diinduksi mutasi dengan iradiasi gamma, (2) Mengetahui adanya interaksi pengaruh penggunaan dosis iradiasi gamma dan konsentrasi zat pengatur tumbuh terhadap perbanyakan tunas pisang cv. Barangan hasil induksi mutasi dengan iradiasi gamma secara in vitro dan (3) Aklimatisasi plantlet dan penyediaan bibit pisang cv. Barangan agar siap ditumbuhkan di lapangan. Percobaan penelitian ini dilaksanakan di laboraturium Kultur Jaringan Tumbuhan, FMIPA, Universitas Negeri Jakarta. Tahapan percobaan meliputi: (1) multiplikasi tunas-tunas pisang barangan hasil iradiasi gamma dan (2) kemampuan tumbuh planlet hasil iradiasi gamma pada saat aklimatisasi. Hasil percobaan tunas pisang cv. Barangan hasil induksi mutasi dengan iradiasi gamma didapatkan dosis letal yang mereduksi pertumbuhan tunas sebesar 50% (LD50) yaitu sebesar 61.54 Gy, terdapat interaksi pengaruh berbagai dosis iradiasi gamma dan berbagai konsentrasi zat pengatur tumbuh terhadap variasi karakter jumlah tunas dan jumlah daun. Jumlah tunas dan jumlah daun tertinggi terdapat pada iradiasi gamma dosis 0 Gy dan yang terendah pada dosis 60 Gy. Percobaan aklimatisasi didapatkan persentase tingkat keberhasilan tanaman yang bertahan hidup sebesar 56% dari total tanaman selama 20 MSA. ************************************************************ Bananas are agricultural food products that have high commercial value and have the opportunity to be developed. The problem faced in banana cultivation is that it is difficult to get quality seeds in large quantities in a short time. The purpose of this study is: (1) to know the letal dose 50% (LD50) to increase the genetic variation of Barangan bananas after induced mutations with gamma irradiation, (2) to know the interaction of the effect of the use of gamma irradiation doses and the concentration of plant growth regulator against the multiplication of banana cv. Barangan shoots. The results of mutation induction by gamma irradiation in vitro and (3) acclimatization of planlets and the provision of banana seedlings cv. Barangan to be ready to be grown in the field. This research experiment was conducted at the Plant Tissue Culture Laboratory, FMIPA, Jakarta State University. The trial stages include: (1) multiplication of banana buds of gamma irradiation and (2) the ability to grow gamma irradiation planlets at the time of acclimatization. The results of mutation induction with gamma irradiation obtained a lethal dose which reduced the growth of shoots by 50% (LD50) which is 61.54 Gy, there is an interaction of the influence of various doses of gamma irradiation and various concentrations of plant growth regulator to variations in the character of the number of shoots and the number of leaves. The highest number of shoots and leaf counts were found at gamma irradiation doses of 0 Gy and the lowest at doses of 60 Gy and acclimatization trials found a percentage of the success rate of surviving plants at 56% of the total plant during 20 MSA.
