Abstract

Rubus elongatus Sm. (Rosaceae), salah satu spesies Rubus yang ditemukan di Indonesia dan dikoleksi di Kebun Raya Cibodas. Spesies Rubus diketahui memiliki beragam senyawa metabolit dengan berbagai aktivitas biologis. Penelitian ini bertujuan untuk menganalisis kandungan metabolit sekunder dan aktivitas antibakteri esktrak etanol dari batang, daun, pucuk, dan bunga R. elongatus. Penelitian ini menggunakan metode deskriptif dan eksperimental dan dianalisis menggunakan SPSS seri 25 dengan ANOVA satu arah dan aktivitas antibakteri dianalisis dengan ANOVA dua arah. Pengambilan sampel diperoleh dari Kebun Raya Cibodas dengan kode I.V. C.201006002. Pengujian dilakukan di laboratorium Badan Riset dan Inovasi Nasional (BRIN). Senyawa fitokimia diidentifikasi menggunakan metode kolorimetri. Kandungan total fenol (TPC) ditentukan dengan metode Follin-Ciocalteu dan dihitung sebagai ekuivalen asam galat, sedangkan kandungan total flavonoid (TFC) ditentukan dengan metode aluminium klorida dan dihitung sebagai ekuivalen kuersetin. Analisis GC-MS digunakan untuk mengidentifikasi senyawa R. elongatus berdasarkan volatilitasnya dan masa molekulnya, dengan identifikasi senyawa berdasarkan NIST20. L. melalui analisis spektra massa GC-MS. Sementara itu, uji aktivitas antibakteri dilakukan dengan menggunakan metode difusi cakram. Analisis fitokimia menunjukkan adanya alkaloid, tanin, fenol, dan flavonoid pada ekstrak etanol R. elongatus. TPC dan TFC ekstrak etanol R. elongatus tertinggi ditemukan pada bunga (20,75 ± 0,12 mg GAE/g dan 17,34 ± 0,69 mg QE/g). Analisis GC-MS mengidentifikasi beberapa senyawa dominan, yaitu Phytol pada daun (50,05%), Bis(2-ethylhexyl) phthalate pada bunga dan pucuk (28,24% dan 16,83%), serta Dibutyl phthalate pada batang (5,80%). Aktivitas antibakteri R. elongatus menunjukkan penghambatan terhadap pertumbuhan S. aureus dengan zona hambat 2,69 mm, namun tidak menunjukkan aktivitas terhadap E. coli dan S. mutans. Penelitian ini mengungkapkan bahwa R. elongatus mengandung berbagai senyawa metabolit dan menunjukkan aktivitas antibakteri. ***** Rubus elongatus Sm. (Rosaceae) is one of the Rubus species found in Indonesia and collected at the Cibodas Botanical Garden. The Rubus species is known to contain various metabolite compounds with diverse biological activities. This study aims to analyze the secondary metabolite content and antibacterial activity of ethanol extract from the stem, leaves, shoots, and flowers of R. elongatus. This study used descriptive and experimental methods and analyzed using SPSS series 25 with one-way ANOVA and antibacterial activity analyzed with two-way ANOVA. Sampling was obtained from Cibodas Botanical Garden with code I.V.C.201006002. Testing was carried out in the laboratory of the National Research and Innovation Agency (BRIN). Phytochemical compounds were identified using colorimetric methods. Total phenol content (TPC) was determined using the Folin-Ciocalteu method and expressed as gallic acid equivalents, while total flavonoid content (TFC) was determined using the aluminum chloride method and expressed as quercetin equivalents. GC-MS analysis was employed to identify R. elongatus compounds based on their volatility and molecular mass, with compound identification using the NIST20.L library through GC-MS mass spectra analysis. Meanwhile, the antibacterial activity test was conducted using the disk diffusion method. Phytochemical analysis revealed the presence of alkaloids, tannins, phenols, and flavonoids in the ethanol extract of R. elongatus. The highest TPC and TFC in the ethanol extract of R. elongatus were found in the flowers (20,75 ± 0,12 mg GAE/g and 17,34 ± 0,69 mg QE/g). GC-MS analysis identified several dominant compounds, including Phytol in the leaves (50,05%), Bis(2-ethylhexyl) phthalate in the flowers and shoots (28,24% and 16,83%), and Dibutyl phthalate in the stem (5,80%). The antibacterial activity of R. elongatus showed inhibition against the growth of S. aureus with a zone of inhibition of 2,69 mm, while no activity was observed against E. coli and S. mutans. This study reveals that R. elongatus contains various metabolite compounds and demonstrates antibacterial activity.
