Abstract
Sel Punca Mesenkim (SPM) adalah tipe sel yang memiliki potensi terapeutik signifikan dan termasuk dalam kategori Advanced Therapy Medicinal Products (ATMP). SPM memiliki kemampuan untuk memperbarui diri secara terus-menerus dan berdiferensiasi menjadi berbagai jenis sel, seperti sel tulang, sel kartilago, dan sel lemak. Kemampuan ini menjadikannya pilihan yang menarik untuk terapi regeneratif dan rekonstruktif. Optimalisasi SPM dilakukan pada prekondisi hipoksia melalui ekspresi Hypoxia-inducible factor 1 alpha. Isolasi SPM bersumber dari tali pusat Macaca fascicularis dan di beri perlakuan prekondisi hipoksia dengan konsentrasi O2 sebesar 3% pada medium kultur free serum, dan prekondisi normoksia 21% pada medium free serum. Data dianalisis secara deskriptif dan dihitung dengan metode livak. Hasil penelitian menunjukan bahwa SPM yang diisolasi dari tali pusat Macaca fascicularis memiliki karakteristik morfologi seperti fibroblast, mengekspresikan marker CD90-FITC, CD105 PerCP-Cy5.5 dan CD73-APC, memiliki sifat poliferasi,diferensiasi, dan mengekspresikan HIF-1α. Hal ini dapat disimpulkan bahwa SPM yang diisolasi dari tali pusat Macaca fascicularis, memenuhi persyaratan untuk digunakan sebagai terapi sel dan prekondisi hipoksia dapat menjadi upaya optimalisasi SPM yang akan digunakan sebagai terapi sel. ***** Mesenchymal stem cells (MSCs) are a cell type that have significant therapeutic potential and fall under the category of Advanced Therapy Medicinal Products (ATMPs). SPM have the ability to continuously self-renew and differentiate into various cell types, such as bone cells, cartilage cells and fat cells. This ability makes them an attractive option for regenerative and reconstructive therapies. Optimization of SPM was performed under hypoxic preconditioning through the expression of Hypoxia-inducible factor 1 alpha. SPM isolation was sourced from the umbilical cord of Macaca fascicularis and treated with hypoxic preconditioning with O2 concentration of 3% in serum-free culture medium, and normoxic preconditioning of 21% in serum-free medium. Data were analyzed descriptively and calculated by the livak method. The results showed that SPM isolated from the umbilical cord of Macaca fascicularis has fibroblast-like morphological characteristics, expresses markers CD90-FITC, CD105 PerCP-Cy5.5 and CD73-APC, has poliferation, differentiation, and expresses HIF-1α. It can be concluded that SPM isolated from the umbilical cord of Macaca fascicularis, meet the requirements to be used as cell therapy and hypoxic preconditioning can be an effort to optimize SPM to be used as cell therapy.
