Abstract
Sapi Bali (Bos javanicus) dikenal sebagai rumpun sapi asli Indonesia yang memiliki keunggulan dalam daya adaptasi, ketahanan terhadap penyakit, dan daging berkualitas dengan rendah lemak. Namun, kemurnian genetiknya terancam oleh perkawinan silang yang tidak terkontrol. Penelitian ini bertujuan untuk mengidentifikasi breed-specific SNP dari gen MC1R pada sapi Bali dan menganalisis keragaman genetiknya pada populasi sapi Bali dengan sampel rumpun sapi lainnya. Penelitian ini menggunakan metode deskriptif kuantitatif dengan menganalisis sampel DNA dari tujuh rumpun sapi sebanyak 140 sampel menggunakan metode PCR-RFLP dengan enzim restriksi NdeI. Data genotipe yang diperoleh kemudian dianalisis untuk mencari nilai frekuensi genotipe dan alel, keseimbangan Hardy-Weinberg (HWE), heterozigositas pengamatan (Ho),heterozigositas harapan (He), dan Fst (fixation index ). Salah satu kandidat SNP yang diduga spesifik pada sapi Bali yaitu SNP AF445641.1:g.3621C>A.Hasil penelitian menunjukkan bahwa didapatkan tiga genotipe(AA,AC, dan CC) pada populasi sapi Bali. Frekuensi alel A (0,67) lebih dominan dibandingkan alel C (0,33). Populasi sapi Bali menunjukkan keragaman genetik yang tinggi dan berada dalam kesetimbangan Hardy-Weinberg. Dapat disimpulkan bahwa SNP AF445641.1:g.3621C>A pada gen MC1R belum dapat digunakan untuk marka genetik tunggal untuk mendeteksi kemurnian sapi Bali. Kata kunci: alel spesifik, kemurnian genetik, marka genetik, sekuensing ***** Balinese cattle (Bos javanicus) is one of the cattle crays native to Indonesia that has advantages in adaptability,resistance to disease, and superior meat. However, its genetic purity is threatened by uncontrolled interbreeding. This study aimed to identify breed-specific SNP of the MC1R gene in Balinese cattle and analyze its genetic diversity in the Balinese cattle population with samples of other cattle herds. This research employed a descriptive quantitative method, analyzing DNA samples from seven cattle breeds, totaling 140 samples, using the PCR-RFLP method with the NdeI restriction enzyme. The obtained genotype data were then analyzed to determine genotype and allele frequencies, Hardy-Weinberg equilibrium (HWE), observed heterozygosity (Ho), expected heterozygosity (He), and Fst (fixation index). One of the candidate SNP suspected to be specific to Balinese cattle is SNP AF445641.1:g.3621C>A. The results showed that three genotypes (AA, AC, and CC) were obtained in the Balinese cattle population. The frequency of allele A (0.67) is more dominant than allele C (0.33). The Balinese cattle population shows high genetic diversity and is in the Hardy-Weinberg equilibrium.It can be concluded that the SNP AF445641.1:g.3621C>A in the MC1R gene cannot yet be used for a single genetic marker to detect the purity of Balinese cattle Keywords: specific allele, genetic purity, genetic marker, sequencing
