Abstract
Hutan gambut di Kalimantan Tengah, khususnya Desa Tumbang Nusa, merupakan salah satu wilayah yang terdampak kebakaran sehingga dilakukan upaya rehabilitasi. Perubahan habitat akibat kebakaran tersebut ditandai oleh perbedaan vegetasi dan ketersediaan sumber pakan yang berpotensi memengaruhi keanekaragaman serangga dan variasi genetik pada serangga yang berhasil bertahan di lingkungan yang berubah. Penelitian ini bertujuan untuk mengidentifikasi jenis serangga dan menganalisis keanekaragaman, variasi genetik, serta hubungan kekerabatan antar individu serangga dominan dengan kerabat dekatnya berdasarkan data sekuen GenBank. Penelitian ini dilakukan menggunakan metode deskriptif kuantitatif melalui identifikasi morfologi hingga tingkat morfospesies dan identifikasi molekuler serangga dominan melalui tahapan ekstraksi DNA, PCR, elektroforesis, dan sekuensing. Analisis komunitas serangga menggunakan indeks keanekaragaman (H’), kemerataan (E), kekayaan jenis (R), dominansi (C), dan indeks kesamaan Bray-Curtis, sedangkan analisis variasi genetik dan filogenetik dilakukan menggunakan DnaSP dan MEGA12. Hasil penelitian menunjukkan bahwa pada hutan alam dan rehabilitasi hutan ditemukan 9 bangsa, 61 suku, dan 101 jenis serangga. Hutan alam memiliki keanekaragaman rendah dengan dominansi tinggi, kekayaan jenis tinggi, dan kemerataan rendah, sedangkan rehabilitasi hutan menunjukkan keanekaragaman sedang dengan dominansi rendah, kekayaan jenis tinggi, dan kemerataan tinggi. Indeks kesamaan kedua habitat tergolong rendah, menunjukkan perbedaan komposisi komunitas serangga. Variasi genetik P. myrmecodiae di hutan alam rendah dan variasi genetik T. minimus di rehabilitasi hutan tergolong tinggi. Secara filogenetik, P. myrmecodiae di Tumbang Nusa berkerabat dekat dengan populasi dari Papua New Guinea dan Myanmar, sedangkan T. minimus berkerabat dekat dengan populasi Thailand yang terdaftar di GenBank. ***** Peat forest in Central Kalimantan, particularly in Tumbang Nusa Village, is among the areas affected by forest fires, prompting rehabilitation efforts. Habitat changes due to fires are characterized by differences in vegetation and food availability, which could potentially affect insect diversity and genetic variation in insects that survived in the changed environment. This study aimed to identify insect species and analyze diversity, genetic variation, and kinship relationships between dominant insects and close relatives based on GenBank sequence data. This study was conducted using quantitative descriptive methods through morphological identification to the morphospecies level and molecular identification of dominant insects through DNA extraction, PCR, electrophoresis, and sequensing. Analysis of insects community used diversity (H’), evenness (E), species richness (R), dominance (C), and Bray-Curtis similarity indices, while genetic variation and phylogenetic relationships were analyzed using DnaSP and MEGA12. Results in natural and rehabilitated forests showed that there were 9 genera, 61 tribes, and 101 species of insect. Natural forests had low diversity with hight dominance, high species richness, and low evenness, while rehabilitated forests showed moderate diversity with low dominance, high species richness, and high evenness. The similarity index of the two habitats was low, indicating differences in insect community composition. Genetic variation of P. myrmecodiae in natural forest was low, while genetic variation of T. minimus in rehabilitated forests was high. Phylogenetically, P. myrmecodiae in Tumbang Nusa was closely related to populations from Papua New Guinea and Myanmar, while T. minimus was closely related to the Thailand population registered in GenBank.
