Abstract

Aeschynanthus mormoratus merupakan tanaman hias dari genus Aeschynanthus yang secara umum dikenal sebagai bunga lipstik dengan bunga khas berbentuk tubular berwarna hijau kekuningan, namun memiliki kapasitas reproduksi yang rendah sehingga perbanyakan secara konvensional kurang optimal. Kultur jaringan menjadi salah satu metode alternatif perbanyakan tanaman ini, dengan pemanfaatan sumber nitrogen organik seperti asam amino glutamin. Penelitian ini bertujuan untuk mengetahui pengaruh penambahan glutamin terhadap perbanyakan tunas dan profil metabolit sekunder A. mormoratus secara in vitro. Metode pada penelitian ini eksperimental dengan desain penelitian Rancangan Acak Lengkap (RAL) non-faktorial. Penelitian ini menggunakan media MS dengan penambahan 1 mg/L TDZ dan 0,5 mg/L. Perlakuan pada penelitian ini meliputi 5 konsentrasi asam amino glutamin yaitu 0, 30, 50, 70, dan 100 mg/L. Eksplan dipanen setelah 6 MSS dengan menggunakan metode ekstraksi maserasi dan pengujian GCMS. Parameter yang diamati meliputi luas area pertumbuhan (cm2), waktu muncul tunas, warna kalus, berat basah (g), berat kering (g), rendemen (%), serta profil metabolit sekunder. Hasil ANOVA menunjukkan bahwa glutamin tidak berpengaruh signifikan (p > 0,05) terhadap luas area pertumbuhan pada tahap inisiasi dan subkultur. Namun, glutamin berpengaruh signifikan (p = 0,001) terhadap berat basah dan berat kering, dengan nilai tertinggi pada perlakuan 70 mg/L. Analisis GCMS mengidentifikasi Ethyl oleate sebagai senyawa dominan dari golongan ester asam lemak pada perlakuan glutamin 100 mg/L dengan persen area sebesar 95,16%. ***** Aeschynanthus mormoratus is an ornamental plant belonging to the genus Aeschynanthus, commonly known as the lipstick plant, characterized by yellowish-green tubular flowers, but it has low reproduction capacity, making conventional propagation less than optimal. Plant tissue culture is one alternative method for propagating this plant, utilizing organic nitrogen sources such as the glutamine. This study aims to determine the effect of glutamine addition on shoot propagation and the secondary metabolite profile of A. mormoratus in vitro. The method used in this study was experimental with a non-factorial Complete Randomized Design (CRD). This study used MS medium with the addition of 1 mg/L TDZ and 0,5 mg/L. The treatments in this study included 5 concentrations of glutamine, namely 0, 30, 50, 70 and 100 mg/L. The explants were harvested after 6 weeks of subculture using the maceration extraction method and GCMS. The parameters observed included growth area (cm), bud emergence time, callus color, fresh weight (g), dry weight (g), yield (%), and secondary metabolite profiles. The ANOVA results showed that glutamine had no significant effect (p > 0,05) on the growth area at the initiation and subculture stages. However, glutamine had a significant effect (p = 0,001) on fresh weight and dry weight, with the highest values in the 70 mg/L treatment. GCMS analysis identified ethyl oleate as the dominant compound of the fatty acid ester group in the 100 mg/L percentage of 95,16%.
