Abstract

Kedelai (Glycine max (L.)) merupakan leguminosa berprotein tinggi yang mengandung fitoestrogen sehingga berpotensi memengaruhi jaringan reproduksi, termasuk folikulogenesis ovarium. Sugentan 2 adalah varietas kedelai hasil iradiasi sinar gamma dengan umur panen sangat genjah (67 hari), namun kandungan fitokimia dan pengaruhnya terhadap ovarium belum diketahui. Penelitian ini bertujuan mengidentifikasi komponen fitokimia ekstrak kedelai Sugentan 2 serta menganalisis pengaruhnya terhadap morfometri dan histologis ovarium mencit (Mus musculus). Penelitian eksperimental ini menggunakan Rancangan Acak Lengkap dengan 25 ekor mencit betina yang dibagi menjadi lima kelompok, yaitu kontrol, kedelai impor dosis 500 dan 1000 mg/kgBB, serta kedelai Sugentan 2 dosis 500 dan 1000 mg/kgBB, yang diberikan secara oral selama 14 hari. Data dianalisis menggunakan ANAVA satu arah yang dilanjutkan uji Beda Nyata Terkecil (BNT) pada taraf 5%. Hasil uji fitokimia kualitatif menunjukkan ekstrak Sugentan 2 mengandung tanin dan triterpenoid dengan intensitas kuat, flavonoid dan saponin dengan intensitas sedang, serta alkaloid dengan intensitas lemah, sedangkan steroid tidak terdeteksi; golongan senyawa ini serupa dengan kedelai impor tetapi dengan intensitas lebih tinggi. Rendemen ekstrak Sugentan 2 sebesar 4,42%, lebih rendah dibandingkan kedelai impor (6,54%). Perlakuan berpengaruh nyata (p<0,05) terhadap panjang dan lebar ovarium, Indeks Gonadosomatik (IGS), jumlah folikel primordial, sekunder, dan atresia, serta diameter folikel primer dan sekunder, tetapi tidak berpengaruh nyata terhadap jumlah folikel primer, folikel de Graaf, korpus luteum, dan diameter folikel de Graaf. Pengaruh kedelai Sugentan 2 bersifat bergantung dosis: dosis 1000 mg/kgBB menghasilkan ovarium terbesar (panjang 3,59 mm; lebar 3,48 mm), jumlah folikel primordial (7,00) dan sekunder (6,40) tertinggi setara kontrol, serta atresia terendah (3,60), namun juga IGS terendah (0,018) dan diameter folikel primer terkecil (0,039 mm); dosis 500 mg/kgBB menghasilkan diameter folikel sekunder tertinggi (0,146 mm). Dengan demikian, kedelai Sugentan 2 dosis 1000 mg/kgBB paling optimal mendukung perkembangan ovarium mencit dengan tingkat atresia yang rendah. ***** Soybean (Glycine max (L.)) is a high-protein legume containing phytoestrogens, compounds capable of influencing reproductive tissues, including ovarian folliculogenesis. Sugentan 2 is a gamma-irradiated soybean cultivar notable for its very short harvest period of 67 days; however, its phytochemical composition and effect on the ovary have not been investigated. This study aimed to identify the phytochemical constituents of Sugentan 2 soybean extract and to analyze its effect on the morphometry and histology of mouse (Mus musculus) ovaries. This experimental study employed a Completely Randomized Design using 25 female mice divided into five groups: control, imported soybean at 500 and 1000 mg/kgBW, and Sugentan 2 soybean at 500 and 1000 mg/kgBW, administered orally for 14 days. Data were analyzed using one-way ANOVA followed by the Least Significant Difference (BNT) test at the 5% level. Qualitative phytochemical screening showed that the Sugentan 2 extract contained tannins and triterpenoids at strong intensity, flavonoids and saponins at moderate intensity, and alkaloids at weak intensity, whereas steroids were not detected; these compound groups were similar to those of imported soybean but with higher intensity. The extract yield of Sugentan 2 was 4.42%, lower than that of imported soybean (6.54%). The treatments significantly affected (p<0.05) ovarian length and width, the Gonadosomatic Index (GSI), the number of primordial, secondary, and atretic follicles, and the diameters of primary and secondary follicles, but had no significant effect on the number of primary follicles, Graafian follicles, corpora lutea, or Graafian follicle diameter. The effect of Sugentan 2 was dose-dependent: the 1000 mg/kgBW dose produced the largest ovaries (length 3.59 mm; width 3.48 mm), the highest primordial (7.00) and secondary (6.40) follicle counts— comparable to the control and the lowest atresia (3.60), yet also the lowest GSI (0.018) and the smallest primary follicle diameter (0.039 mm); the 500 mg/kgBW dose produced the largest secondary follicle diameter (0.146 mm). Thus, Sugentan 2 soybean at 1000 mg/kgBW was the most optimal in supporting mouse ovarian development with a low level of atresia.
