Abstract
Nangkadak super O merupakan varietas baru hasil persilangan antara dua spesies dari genus Artocarpus yaitu Artocarpus heterophyllus (nangka) dan Artocarpus integer (cempedak). Genus Artocarpus diketahui kaya akan kandungan fenolik seperti flavonoid, stilbenoid dan arilbenzofuran yang berpotensi digunakan sebagai antioksidan, antibakteri, antidiabetes, antimalaria dan antiinflamasi. Penelitian mengenai kandungan metabolit sekunder tanaman pada genus Artocarpus khususnya Artocarpus heterophyllus dan Artocarpus integer telah banyak dilakukan, namun belum ada informasi penelitian mengenai kandungan metabolit sekunder Nangkadak super O. Penelitian ini bertujuan untuk mengisolasi, mengidenifikasi dan menguji aktivitas antioksidan metabolit sekunder dari fraksi kloroform kayu batang Nangkadak Super O. Proses isolasi dilakukan melalui beberapa tahap diantaranya ekstraksi, fraksinasi dengan metode kromatografi vakum cair dan pemurnian dengan metode kromatografi radial. Pembuktian senyawa murni dilakukan dengan uji tiga sistem kepolaran dan penyemprotan reagen spesifik masing-masing dengan noda tunggal. Senyawa hasil isolasi kemudian diidentifikasi strukturnya dengan analisis data spektrum UV-vis, FTIR, 1H-NMR,13C-NMR, HSQC dan HMBC, serta dilakukan uji aktivitas antioksidan dengan menggunakan metode DPPH. Hasil penelitian diperoleh dua isolat murni yang teridentifikasi sebagai siklomorusin dan sikloartokarpin berdasarkan analisis data spektrum UV-vis, FTIR, 1H-NMR,13C-NMR, HSQC dan HMBC. Hasil uji antioksidan menunjukkan bahwa siklomorusin (29) dan sikloartokarpin (31) memiliki aktivitas antioksidan lemah yang ditunjukkan dengan nilai IC50 yaitu 166 dan 158,5 ppm ************* Nangkadak super O is a new variety of hybrid between Artocarpus heterophyllus (nangka) and Artocarpus integer(cempedak). Genus Artocarpus are rich in phenolic compounds including flavonoids, stilbenoids, and arylbenzofurons which are potentially used as antioxidant, antibacterial, antidiabetic, antimalarial and antiinflamatory. There have been many reports on secondary metabolite researchs on Genus Artocarpus,especially Artocarpus heterophyllusand Artocarpus integer, however no information is documented on Nangkadak super O researchs. The purposes of this research were toisolate, identify and antioxidant activitytestsof secondary metabolite from chloroform extract of Nangkadak Super O stems. Isolation was carried out in several steps: extraction, fractionation by vacuum liquid chromatography and purification by radial chromatography. Proof of pure compounds were carried out by three systems of polarity and spraying with spesific reagent. The isolated compounds identified by data analysis UV-vis, FTIR, 1H-NMR,13C-NMR, HSQC and HMBC, and an antioxidant activity tests were carried out using DPPH method. Theresults were obtained 2 isolates were identified as cyclomorusin and cycloartocarpin based on data analysis UV-vis, FTIR, 1H-NMR,13C-NMR, HSQC and HMBC. The result of antioxidant activity test show that Cyclomorusin (29)and Cyloartocarpin(31) have weak antioxidant activity as indicated by IC50values of166and 158,5ppm.
