Abstract

Temu kunci merupakan tanaman rempah yang mengandung metabolit sekunder yang dapat digunakan sebagai bahan baku obat. Mikroorganisme yang terkandung dalam jaringan tanaman temu kunci dapat diisolasi untuk mendapatkan kapang Dark Septate Endophytes (DSE). Penelitian ini bertujuan untuk menguji kemampuan kapang asal temu kunci yaitu Byssochlamys spectabilis UNJCC F2 dan Pleosporales sp. UNJCC sebagai antimikroba dan antioksidan. Desain penelitian menggunakan Rancangan Acak Lengkap (RAL). Data dianalisis menggunakan ANAVA dua arah kemudian dilanjutkan uji Duncan Multiple Range Test (DMRT) 5%. Tahapan penelitian terdiri dari, pengamatan morfologi kapang DSE, kurva pertumbuhan kapang, fermentasi kapang, uji aktivitas antimikroba terhadap bakteri Listeria monocytogenes, Salmonella enteritidis, dan khamir patogen Candida albicans, dan uji aktivitas antioksidan yang meliputi uji analisis total flavonoid dengan spektrofotometer, penentuan kurva standar vitamin C, penentuan kadar flavonoid total, uji aktivitas antioksidan dengan metode DPPH dengan variasi konsentrasi 1.875 ppm, 3.125 ppm, dan 5.000 ppm. Hasil pengujian aktivitas antimikroba kapang B. spectabilis UNJCC F2 dan Pleosporales sp. UNJCC F6 menghasilkan zona hambat terhadap bakteri S. enteritidis sebesar 3,63 mm dan 5,18 mm, kapang B. spectabilis UNJCC F2 dan Pleosporales sp. UNJCC F6 terhadap L. monocytogenes sebesar 4,65 mm dan 4,58 mm, dan kapang B. spectabilis UNJCC F2 dan Pleosporales sp. UNJCC F6 terhadap C. albicans tidak menghasilkan zona hambat. Hasil pengujian aktivitas antioksidan dengan metode DPPH menunjukkan bahwa kapang B. spectabilis UNJCC F2 menghasilkan nilai persentase inhibisi tertinggi yaitu sebesar 62,32% pada konsentrasi 5.000 ppm dan pada Pleosporales sp. UNJCC F6 menghasilkan nilai persentase inhibisi tertinggi sebesar 22,52% pada konsentrasi 5000 ppm. Berdasarkan hasil penelitian, kapang Pleosporales sp. UNJCC F6 memiliki kemampuan terbaik dalam menghambat bakteri patogen dan B. spectabilis UNJCC F2 memiliki kemampuan sebagai antioksidan yang baik. Temu Kunci is a spice plant that contains secondary metabolites that can be used as medicinal raw materials. Microorganisms contained in the plant tissue of Intersection can be isolated to obtain Dark Septate Endophytes (DSE) molds. This study aimed to test the ability temu kunci of fungi, namely Byssochlamys spectabilis UNJCC F2 and Pleosporales sp. UNJCC as antimicrobial and antioxidant. The research design used a completely randomized design (CRD). The data were analyzed using two-way ANOVA and then continued with the 5% Duncan Multiple Range Test (DMRT). The research stages consisted of observing the morphology of the DSE mold, mold growth curve, mold fermentation, antimicrobial activity test against the bacteria Listeria monocytogenes, Salmonella enteritidis, and the yeast pathogenic Candida albicans, and an antioxidant activity test which included an analysis test of total flavonoids with a spectrophotometer, determination of the standard curve vitamin C, determination of total flavonoid levels, antioxidant activity test using the DPPH method with various concentrations of 1.875 ppm, 3.125 ppm, and 5.000 ppm. The results of testing the antimicrobial activity of B. spectabilis UNJCC F2 and Pleosporales sp. UNJCC F6 produced 3,63 mm and 5.18 mm inhibition zones against S. enteritidis bacteria, the mold B. spectabilis UNJCC F2 and Pleosporales sp. UNJCC F6 against L. monocytogenes at 4,65 mm and 4,58 mm, and molds B. spectabilis UNJCC F2 and Pleosporales sp. UNJCC F6 against C. albicans did not produce inhibition zones. The results of the antioxidant activity test using the DPPH method showed that the mold B. spectabilis UNJCC F2 produced the highest inhibition value which was 62,32% at a concentration of 5.000 ppm and mold Pleosporales sp. UNJCC F6 produced the highest inhibition value which was 22,52% at a concentration of 5.000 ppm. Based on the research results, the fungus Pleosporales sp. UNJCC F6 has the best ability to inhibit pathogenic bacteria and B. spectabilis UNJCC F2 has the ability as a good antioxidant.
