Abstract

Mikroplastik didefinisikan sebagai semua partikel plastik dengan diameter kurang dari 5 mm. Mikroplastik polipropilena (PP) memiliki sifat rekalsitran sehingga sulit terdegradasi dan dapat bertahan di alam dalam waktu yang cukup lama. Metode remediasi mikroplastik oleh bakteri menjadi solusi potensial untuk mengatasi pencemaran mikroplastik di lingkungan. Tujuan penelitian ini yaitu untuk mendeteksi serta mengetahui kemampuan bakteri pendegradasi mikroplastik PP asal limbah plastik Teluk Jakarta yang diperoleh dari stasiun berbeda, yaitu Muara Kamal (Stasiun 1), Muara Angke (Stasiun 2), dan Marina (Stasiun 3). Tahapan penelitian ini terdiri dari kultur pengayaan bakteri asal sampel limbah plastik Teluk Jakarta pada media Zobell marine broth dan uji degradasi mikroplastik PP pada Mineral Salt Medium (MSM) selama 60 hari menggunakan inokulum bakteri hasil kultur pengayaan. Parameter yang diamati meliputi nilai OD dan jumlah koloni hasil kultur pengayaan, serta nilai pH akhir media uji, berat kering mikroplastik, dan analisis kerusakan senyawa polipropilena menggunakan spektroskopi FTIR hasil uji degradasi mikroplastik PP oleh bakteri. Berdasarkan hasil yang diperoleh, bakteri yang berasal dari ketiga stasiun dapat mendegradasi mikroplastik PP. Hasil ANAVA satu arah menunjukkan bakteri yang berasal dari stasiun berbeda, memberikan pengaruh nyata terhadap pengurangan mikroplastik PP dengan pengurangan berat kering berkisar 2,16-6,6 %. Berdasarkan uji DMRT 5%, bakteri asal stasiun 2 menghasilkan tingkat degradasi mikroplastik tertinggi dengan pengurangan berat kering sebesar 6,6 %. Hasil penelitian ini diharapkan dapat memberikan informasi dasar bakteri pendegradasi mikroplastik polipropilena asal Teluk Jakarta. Kata Kunci. Polypropylene (PP) microplastics have recalcitrant properties, making them difficult to degrade and can last in nature for quite a long time. The method of remediation of microplastics by bacteria is a potential solution to overcome microplastic pollution in the environment. The purpose of this study was to detect and determine the ability of PP microplastic degrading bacteria from Jakarta Bay plastic waste obtained from different stations, namely Muara Kamal (Station 1), Muara Angke (Station 2), and Marina (Station 3). The stages of this research included of enrichment culture of bacteria from plastic waste samples in Jakarta Bay on Zobell marine broth media and microplastic PP degradation tests on Mineral Salt Medium (MSM) for 60 days using bacterial inoculum from enrichment culture. Parameters observed included the optical density (OD) value and the number of colonies resulting from the enrichment culture, as well as the final pH value of the MSM medium, the dry weight of the microplastics, and analysis of the breakdown of polypropylene compounds using FTIR spectroscopy as a result of the degradation test of PP microplastics by bacteria. Based on the results obtained, bacteria from the three stations could degrade PP microplastics. The results of a one-way ANOVA showed that bacteria originating from different stations had a significant effect on reducing microplastic PP with a reduction in dry weight ranging from 2,16 to 6,6%. Based on the 5% DMRT test, bacteria from station 2 produced the highest level of microplastic degradation with a dry weight reduction of 6.6%. The results of this study are expected to provide basic information on polypropylene microplastic degrading bacteria from Jakarta Bay. Keywords.
