Abstract
Infeksi menular seksual (IMS) masih menjadi salah satu ancaman serius bagi umat manusia, salah satunya adalah IMS yang disebabkan oleh patogen gonore. Berdasarkan data Kementerian Kesehatan Republik Indonesia mencapai 2486 orang baru terinfeksi gonore di Indonesia. Gonore disebabkan oleh bakteri Neisseria gonorrhoeae yang hanya mampu bertempat di dalam tubuh manusia. Deteksi cepat dan akurat menjadi salah satu upaya yang diperlukan dalam menangani infeksi bakteri N. gonorrhoeae. Penelitian ini bertujuan untuk mendeteksi keberadaan N. gonorrhoeae pada sampel usap vagina wanita pekerja seks di Indonesia menggunakan metode RT-PCR berdasarkan pada pseudogen porA dan resistensi N. gonorrhoeae terhadap antibiotik azitromisin melalui metode RT-PCR berdasarkan gen 23S rRNA. Metode penelitian menggunakan metode deskriptif dengan jumlah sampel sebanyak 84 BBT (Bahan Biologi Tersimpan) dari hasil usap vagina. Data yang diperoleh, dianalisa secara deskriptif menggunakan program komputer Bio-rad CFX manager software. Hasil penelitian menunjukkan bahwa dari 84 sampel, sebanyak 15 sampel (17,86%) terdeteksi negatif N. gonorrhoeae dan 69 sampel (82,14%) positif terinfeksi N. gonorrhoeae. Hasil ini menunjukkan bahwa pseudogen porA merupakan target yang baik untuk mendeteksi N. gonorrhoeae. Hasil uji resistensi memperlihatkan semua N. gonorrhoeae yang terdeteksi tidak mengalami resistensi azitromisin. Hasil penelitian ini memberikan informasi bahwa terapi infeksi N. gonorrhoeae dengan azitromisin masih efektif dilakukan pada pasien terinfeksi N. gonorrhoeae. ***** Sexually transmitted infections (STIs) are still a serious threat to humanity, one of which is an STI caused by the pathogen gonorrhea. Based on data from the Ministry of Health of the Republic of Indonesia, there were 2,486 new people infected with gonorrhea in Indonesia. Gonorrhea is caused by the bacterium Neisseria gonorrhoeae which can only live in the human body. Fast and accurate detection is one of the efforts needed in dealing with N. gonorrhoeae bacterial infection. This study aims to detect the presence of N. gonorrhoeae in vaginal swab samples of female sex workers in Indonesia using the RT-PCR method based on the porA pseudogene and the resistance of N. gonorrhoeae to the antibiotic azithromycin through the RT-PCR method based on the 23S rRNA gene. The research method uses a descriptive method with a total sample of 84 BBT (Stored Biological Materials) from vaginal swabs. The data obtained were analyzed descriptively using the Bio-rad CFX manager software computer program. The results showed that of the 84 samples, 15 samples (17.86%) were negative for N. gonorrhoeae and 69 samples (82.14%) were positive for N. gonorrhoeae. These results indicate that the porA pseudogene is a good target for detecting N. gonorrhoeae. Resistance test results showed that all detected N. gonorrhoeae did not experience azithromycin resistance. The results of this study provide information that the treatment of N. gonorrhoeae infection with azithromycin is still effective in patients infected with N. gonorrhoeae.
