Abstract

Intensitas penggunaan plastik yang tinggi dan tidak adanya pengelolaan pasca penggunaan yang baik, dapat membuat plastik menjadi salah satu bahan yang mencemari lingkungan. Bioremediasi oleh mikroorganisme dapat digunakan untuk mengendalikan permasalahan tersebut. Tujuan penelitian ini yaitu untuk mendapatkan serta mengetahui kemampuan aktinomisetes pendegradasi plastik LDPE asal sedimen mangrove kota Balikpapan dan Samarinda ditinjau dari pertumbuhan aktinomisetes, nilai pH, berat kering plastik serta struktur kimia dan fisik plastik. Tahapan penelitian ini terdiri dari isolasi pada media AIA 10%, penapisan dengan media Bushnell Haas (BH) yang diperkaya PEG, dan uji degradasi plastik LDPE pada media BH selama 40 hari. Parameter yang diamati meliputi nilai OD, pH, persentase kehilangan berat kering, analisis kerusakan gugus fungsi LDPE menggunakan spektroskopi FTIR, dan analisis morfologi permukaan LDPE dengan SEM. Sebanyak 33 isolat berhasil diisolasi dari sedimen mangrove. Hasil penapisan diperoleh isolat ACTKT22-19, ACTKT22-14, dan ACTKT22-17 memiliki indeks zona bening terbesar. Berdasarkan hasil ANOVA dua arah dan uji DMRT 5%, isolat ACTKT22-19, ACTKT22-14, dan ACTKT22-17, dan konsorsium memiliki kemampuan yang serupa dan perlakuan waktu memberikan pengaruh nyata terhadap persentase kehilangan berat kering dengan pengurangan berat kering sebesar 0,418 ± 0,03% pada hari ke-40 terhadap persentase kehilangan berat kering pada hari ke-10. Konsorsium memiliki nilai pH akhir tertinggi sebesar 7,377 ± 0,05 dan nilai OD tertinggi pada fase log sebesar 0,118 ± 0,02. Hasil FTIR menunjukkan kenaikan nilai transmitansi pada semua perlakuan dan ditemukan kerusakan berupa lubang pada pengamatan permukaan plastik. Hasil penelitian ini diharapkan dapat memberikan informasi dasar aktinomisetes pendegradasi plastik LDPE asal sedimen mangrove Kalimantan Timur. ***** The high intensity of plastic use and the absence of good post-use management can make plastic one of the materials that pollute the environment. LDPE (low-density-polyethylene) is one type of plastic that is often used in everyday life as plastic bags and food wrappers. One of the efforts to control these problems is the bioremediation method by microorganisms. The purpose of this research is to detect and determine the ability of LDPE plastic degrading actinomycetes from East Kalimantan mangrove sediments obtained from different cities, namely Balikpapan and Samarinda. The stages of this research consisted of isolation on 10% AIA media, screening with PEG-enriched Bushnell Haas (BH) media, and LDPE plastic degradation test on BH for 40 days. Parameters observed included OD value, pH, percentage of dry weight loss, damage analysis of LDPE functional groups using FTIR spectroscopy, and surface morphology test of LDPE with SEM. A total of 33 isolates were successfully isolated from mangrove sediments. The screening results obtained isolates ACTKT22-19, ACTKT22-14, and ACTKT22-17 have the largest clear zone index. Based on the results of two-way ANOVA and 5% DMRT test, it showed that ACTKT22-19, ACTKT22-14, and ACTKT22-17, and the consortium had similar abilities and time treatment had a significant effect on the percentage of dry weight loss with a dry weight reduction of 0,418 ± 0,03% on day 40. The consortium had the highest final pH value of 7.377 ± 0.05 and the highest OD value in the log phase of 0,118 ± 0,02. FTIR results showed an increase in transmittance values in all treatments and damage in the form of holes was found in the observation of the plastic surface. The results of this study are expected to provide basic information on LDPE plastic degrading actinomycetes from East Kalimantan mangrove sediments.
