Abstract

Protease merupakan enzim yang mengkatalisasi pemecahan ikatan peptida dalam protein. Enzim tersebut memiliki nilai ekonomi yang tinggi, karena penggunaannya yang luas diberbagai bidang industri. Tujuan penelitian ini untuk mengetahui jenis bakteri yang diisolasi dari tanah asal Kawasan Taman Nasional Gunung Merapi serta mengetahui aktivitas proteasenya pada pengaruh pH. Metode yang digunakan dalam penelitian ini adalah deskriptif meliputi tahapan skrining dan identifikasi isolat bakteri proteolitik, dan metode eksperimental dilakukan pada tahap uji aktivitas protease pada variasi pH 5,5; 6,0; 6,5; 7,0; 7,5; dan 8,0, menggunakan rancangan acak lengkap. Tahapan penelitian meliputi skrining berdasarkan indeks proteolitik, karakterisasi morfologi dan biokimia, identifikasi molekuler, dan uji aktivitas protease dengan pengaruh pH yang berbeda. Berdasarkan hasil pengukuran indeks proteolitik, diperoleh tiga bakteri yang memiliki nilai indeks terbesar, yaitu TKK1A, TKK3B, dan TN2A. Hasil karakterisasi ketiga bakteri merupakan Gram positif. Analisis uji biokimia isolat TKK1A bereaksi positif pada uji VP dan oksidasi, isolat TKK3B dan TN2A bereaksi negatif pada uji indol, metil merah, VP, sitrat, oksidasi. Analisis sekuen 16S rRNA menunjukkan bahwa isolat TKK1A, TKK3B, dan TN2A secara berurut teridentifikasi sebagai Bacillus tequilensis (99,62%), Priestia aryabhattai (100%), Priestia megaterium (99,92%). Hasil ANAVA dua arah menunjukkan tidak adanya pengaruh nyata jenis isolat terhadap aktivitas protease, namun pada pH menunjukkan adanya pengaruh nyata terhadap aktivitas protease yang dihasilkan. Berdasarkan uji DMRT 5%, aktivitas protease tertinggi terdapat pada pH 8 yaitu sebesar 0,0318 U/mL dan terendah pada pH 6,5 sebesar 0,0256 U/mL. Jenis bakteri yang diperoleh pada penelitian ini menghasilkan enzim protease yang bersifat alkali dengan pH optimum 8, dan dapat digunakan sebagai alternatif pengganti bahan kimia untuk diaplikasikan pada berbagai bidang industri.*****Proteases are enzymes that catalyze the breakdown of peptide bonds in proteins. This enzyme has a high economic value, because of its wide use in various industrial fields. The purpose of this study was to determine the type of bacteria isolated from the soil of origin of Mount Merapi National Park and to determine their protease activity on the influence of pH. The method used in this study was descriptive which included screening and identification of proteolytic bacteria isolates, and the experimental method was carried out at the protease activity test stage at a variation of pH 5.5; 6.0; 6.5; 7.0; 7.5; and 8.0, using a completely randomized design. The stages of the research included screening based on proteolytic index, morphological and biochemical characterization, molecular identification, and testing of protease activity with different pH influences. Based on the results of measuring the proteolytic index, three bacteria with the highest index values were obtained, namely TKK1A, TKK3B, and TN2A. The results of the characterization of the three bacteria were Gram-positive. Biochemical test analysis of TKK1A isolates reacted positive to the VP and oxidation tests, and TKK3B and TN2A isolates reacted negative to the indole, methyl red, VP, citrate, and oxidation tests. 16S rRNA sequence analysis showed that isolates TKK1A, TKK3B, and TN2A were identified as Bacillus tequilensis (99.62%), Priestia aryabhattai (100%), and Priestia megaterium (99.92%) respectively. Two-way ANOVA results showed no significant effect of the type of isolate on protease activity, but the pH showed a significant effect on the resulting protease activity. Based on the 5% DMRT test, the highest protease activity was at pH 8 which was 0.0318 U/mL and the lowest at pH 6.5 was 0.0256 U/mL. The types of bacteria obtained in this study produce protease enzymes that are alkaline have an optimum pH of 8, and can be used as an alternative to chemicals for application in various industrial fields.
