Abstract

Penyakit bacterial spot (BS) yang disebabkan Xanthomonas campestris pv. vesicatoria merupakan penyakit yang menginfeksi tanaman tomat. Pengendalian penyakit ini dapat dilakukan menggunakan pengendali hayati salah satunya bakteri endofit Pseudomonas lactis. Penelitian ini bertujuan untuk menguji kemampuan P. lactis dalam menekan patogen secara in vitro, menguji reaksi hipersensitivitas P. lactis pada tanaman tomat, menguji timgkat patogenisitas X. campestris pv. vesicatoria pada tanaman tomat, menguji efektivitas P. lactis dalam mengontrol penyakit bacterial spot pada tanaman tomat. P. lactis yang digunakan sebanyak 3 strain yang diperoleh dari koleksi BBUSKP. Parameter yang diamati berupa zona bening, keterjadian penyakit, keparahan penyakit, dan pertumbuhan tanaman tomat. Metode yang digunakan adalah deskriptif dan eksperimental dengan desain Rancangan Acak Lengkap (RAL). Hasil uji antagonisme diperoleh P. lactis EB6 menghasilkan zona bening (12,15 ± 1,26) dan indeks hambatan (0,49 ± 0,13) terbesar dibandingkan strain EA1 dan EB4. P. lactis EA1, EB4, dan EB6 tidak memunculkan reaksi hipersensitivitas pada tanaman tomat sehingga dapat digunakan sebagai biokontrol. Hasil uji patogenisitas X. campestris pv. vesicatoria pada tanaman tomat usia 6 MST menunjukkan konsentrasi bakteri 109 cfu/ml memberikan persentase tertinggi (79,16%) pada kedua varietas tomat. P. lactis EB6 menunjukkan efektivitas tertinggi (87,09%) dalam mengontrol bacterial spot dan menurunkan keterjadian penyakit sebesar 75% pada varietas Intan dengan metode penyemprotan. P. lactis EB6 secara umum memberikan hasil terbaik pada peningkatan pertumbuhan tanaman tomat. Bacterial spot disease (BS) caused by Xanthomonas campestris pv. vesicatoria is a disease that infects tomato plants. Control of this disease can be done using biological control, one of which is the endophytic bacterium Pseudomonas lactis. This study aims to test the ability of P. lactis in suppressing pathogens in vitro, testing the hypersensitivity reaction of P. lactis on tomato plants, testing the pathogenicity of X. campestris pv. vesicatoria on tomato plants, testing the effectiveness of P. lactis in controlling bacterial spot disease on tomato plants The P. lactis used were 3 strains obtained from the BBUSKP collection. Parameters observed were clear zone, disease occurrence, disease severity, and tomato plant growth. The method used was descriptive and experimental with a completely randomized design. The results of the antagonism test obtained by P. lactis EB6 produced the largest clear zone (12.15 ± 1.26) and inhibition index (0.49 ± 0.13) compared to strains EA1 and EB4. P. lactis EA1, EB4, and EB6 did not cause hypersensitivity reactions in tomato plants so that it can be used as a biocontrol. The pathogenicity test results of X. campestris pv. vesicatoria on tomato plants aged 6 weeks after planting showed that the concentration of 109 cfu/ml bacteria gave the highest percentage (79.16%) in both tomato varieties. P. lactis EB6 showed the highest effectiveness (87.09%) in controlling bacterial spot and reducing disease occurrence by 75% in the Intan variety with the spraying method. P. lactis EB6 generally gave the best results in terms of bacterial spot control.
