Abstract

Rimpang jahe merah (Zingiber officinale Roscoe), kencur (Kaempferia galanga L.) dan temulawak (Curcuma zanthorrhiza Roxb.) merupakan tanaman obat yang memiliki kandungan senyawa metabolit sekunder berupa fenolik dan flavonoid yang dapat bermanfaat sebagai antioksidan. Tujuan pada penelitian ini yaitu untuk mengetahui profil senyawa fenolik dan flavonoid secara kualitatif dan kuantitatif, serta aktivitas antioksidan kombinasi ekstrak rimpang jahe merah, kencur, dan temulawak melalui metode DPPH, ABTS, dan FRAP. Metode penelitian yang digunakan adalah metode deskriptif dan eksperimental. Metode deskriptif digunakan untuk skrining senyawa fenolik dan flavonoid dari kombinasi ekstrak rimpang jahe merah, kencur, dan temulawak. Metode eksperimental digunakan untuk menganalisis kadar total fenolik dan total flavonoid, serta aktivitas antioksidan melalui metode DPPH, ABTS, dan FRAP. Desain penelitian menggunakan Rancangan Acak Lengkap (RAL) faktorial dengan dua faktor yaitu variasi formulasi kombinasi ekstrak rimpang jahe merah, kencur, dan temulawak (A: 1:1:1, B: 1:1:2, C: 1:2:1, dan D: 2:1:1) dan konsentrasi ekstrak (10, 50, 100, 150, dan 200 ppm). Hasil penelitian ini didapatkan semua kombinasi positif mengandung senyawa fenolik dan flavonoid dengan kadar total fenolik dan total flavonoid tertinggi terdapat pada kombinasi D (perbandingan jahe merah : kencur : temulawak sebesar 2:1:1) dengan konsentrasi 200 ppm secara berurutan yaitu sebesar 370,04 mgGAE/g dan 131,13 mgQE/g. Lalu, formulasi optimum kombinasi ekstrak didapatkan pada kombinasi ekstrak D (2:1:1) dengan nilai IC50 menggunakan metode ABTS sebesar 21,25 ppm. ***** Red ginger (Zingiber officinale Roscoe), Kencur (Kaempferia galanga L.) and Curcuma (Curcuma zanthorrhiza Roxb.) rhizome are medicinal plants that contain secondary metabolite compounds in the form of phenols and flavonoids that can be useful as antioxidants. The purpose of this study was to determine the profile of phenolic and flavonoid compounds qualitatively and quantitatively, the antioxidant activity of the combination of red ginger, kencur, and curcuma extracts based on the DPPH, ABTS, and FRAP methods. The research methods used were descriptive and experimental methods. The descriptive method was used to screen the phenolic and flavonoid compounds from the combination of extracts. The experimental method was used to analyze the total phenolic and total flavonoid levels, and antioxidant activity based on the DPPH, ABTS, and FRAP methods. The research design used a Completely Randomized Design (CRD) with two factorials are variations combination formulations of red ginger : kencur : curcuma (A: 1:1:1, B: 1:1:2, C: 1:2:1, and D: 2:1:1) and extract concentrations (10, 50, 100, 150, and 200 ppm). The results of this study showed that all of combination positive contains phenolic and flavonoid compound and the highest total phenolic and total flavonoid levels were found in combination D (ratio of red ginger : kencur : curcuma are 2:1:1) with a concentration of 200 ppm, are 370.04 mgGAE/g and 131.13 mgQE/g. Then, the optimal formula combination of extracts obtained was a combination of extract D at a ratio of 2:1:1 with an IC50 value using the ABTS method of 21.25 ppm.
