Abstract

This study focuses on the development of a Surface-Enhanced Raman Scattering (SERS) substrate based on gold nanoparticles (AuNPs) to enhance the sensitivity of methylene blue (MB) detection. The AuNPs were synthesized via the reduction of HAuCl₄ using HEPES as both the reducing and stabilizing agent under ultraviolet (UV) irradiation. Variations in HEPES concentration showed that 0.1 M produced nanoparticles with the best optical characteristics based on UV-Vis spectroscopy results. TEM characterization confirmed the formation of gold nanoplate structures, while X-Ray Diffraction (XRD) analysis indicated a dominant peak at the (111) plane, supporting the presence of anisotropic morphology. The SERS substrate was prepared by the drop casting method onto a silicon wafer surface and tested with MB solutions at six different concentrations (10 ppm; 5 ppm; 0.1 ppm; 0.01 ppm; 0.001 ppm; 0.0001 ppm). Raman measurements revealed that the substrate could detect methylene blue down to a concentration of 0.0001 ppm, with a significant increase in spectral intensity compared to Raman measurements without the substrate. A specific achievement of this study is the successful fabrication of a highly sensitive AuNP-based SERS substrate capable of enhancing the detection of dye molecules at ultra-low concentrations.
