Abstract
Tanaman resisten layu Fusarium dapat dikembangkan dengan mutasi dan teknik seleksi in vitro. Tujuan penelitian ini adalah untuk mendapatkan tanaman pisang cv. Ampyang (Musa acuminata, AAA) resisten terhadap layu Fusarium. Metode penelitian yang digunakan yaitu deskriptif. Penelitian dilakukan pada bulan Desember 2013 - November 2014 melalui tiga tahap yaitu multiplikasi tunas, aklimatisasi, dan evaluasi pertumbuhan tanaman pisang di rumah kaca. Bahan tanaman yang digunakan dalam percobaan ini adalah 9 (sembilan) kode klon pisang cv. Ampyang hasil induksi mutasi iradiasi sinar gamma (30, 45, dan 50 Gy) dan seleksi in vitro di rumah kaca koleksi Laboratorium Kultur Jaringan Tanaman - Biologi FMIPA UNJ. Planlet dimultiplikasi selama empat bulan pada media Murashige dan Skoog (MS) dengan 2.25 mg L-1 BAP + 0.22 mg L-1 TDZ + 0.175 mg L-1 IAA, menghasilkan 644 tunas dari ke-9 klon. Evaluasi variasi tanaman pisang usia 6 bulan setelah diaklimatisasi memperlihatkan adanya keragaman fenotipik pada karakter kuantitatif dan kualitatif tanaman. Evaluasi di rumah kaca untuk karakter kuantitatif diperoleh 159 tanaman dari 270 tanaman, yang diaklimatisasi, yang mampu bertahan hidup sampai usia 7 bulan. Tanaman yang mampu bertahan hidup pada saat evaluasi di rumah kaca diidentifikasi sebagai tanaman putatif resisten terhadap layu Fusarium. Plants resistance Fusarium wilt can be developed by mutation and selection in vitro techniques. The purpose of this study was to obtain banana plants cv. Ampyang (Musa acuminata, AAA) resistance to Fusarium wilt. The method used descriptive. The study was conducted in December 2013 - November 2014 through three stages: shoot multiplication, acclimatization, and evaluation of banana plants growing in the green house. The plant material this study was 9 (nine) code clones banana cv. Ampyang result of mutations induced gamma ray irradiation (30, 45, and 50 Gy) and in vitro selection in the green house collection Plant Tissue Culture Laboratory - Biological Science UNJ. Plantlets multiplied for four months on Murashige and Skoog (MS) with 2.25 mg L-1 BAP + 0.22 mg L-1 TDZ + 0.175 mg L-1 IAA, generated 644 shoots from 9th clones. Evaluation of banana plant variation age of 6 months after acclimatized showed phenotypic diversity in quantitative and qualitative character of the plant. Evaluation in the green house for quantitative trait acquired 159 of 270, plants were acclimatized, survived until the age of 7 months. Plants are able to survive at the time of evaluation in the green house are identified as plants that have resistance to Fusarium wilt.
