Abstract
*ABSTRAK* Konsumsi minyak trans yang berlebih dapat mengganggu sistem pencernaan sehingga absorbsi nutrient terhambat. VCO mengandung asam lemak jenuh yang berperan mengikat radikal bebas dan antioksidan yang baik. Perlu penelitian lebih lanjut apakah VCO mampu memperbaiki saluran pencernaan yang rusak akibat konsumsi minyak trans berlebih. Penelitian ini bertujuan mengetahui pengaruh VCO terhadap tinggi vili dan tebal mukosa duodenum pada mencit yang diinduksi minyak trans (MT) dosis tinggi. Sebanyak 25 ekor mencit jantan strain DDY berumur 2 bulan dibagi menjadi 5 kelompok yaitu Kontrol (-) NaCl 0,9%; Kontrol (+) MT; K1 (MT+VCO 200 m g/KgBB); K2 (MT+VCO 400 m g/KgBB); K3 (MT+VCO 600 m g/KgBB). Setelah 8 minggu perlakuan, duodenum diambil dan dibuat preparat histologis. Tinggi vili dan mukosa duodenum diukur dengan software Image-J pada lima lapang pandang. Data diuji secara statistik menggunakan ANOVA pada α=0,05 dan dilanjutkan dengan uji Duncan. Morfologi mukosa duodenum dianalisis secara deskriptif. Hasil pengukuran tinggi vili mukosa duodenum kelompok yang diberi MT+VCO 200 mg/KgBB sebesar 517,37±21,78 µm, MT+VCO 400 mg/KgBB sebesar 574,9±26,61 µm dan MT+VCO 600 mg/KgBB sebesar 651,1±41,74 µm. Hasil pengukuran tebal mukosa duodenum yang diberi MT+VCO 200 mg/KgBB sebesar 525,61, MT+VCO 400 mg/KgBB sebesar 556,6±20,22 µm dan MT+VCO 600 mg/KgBB sebesar 614,2±29,20 µm. Pengukuran tinggi vili duodenum dan tebal mukosa duodenum berbeda nyata dengan kelompok MT (Sig <0.05). Struktur lapisan mukosa duodenum pada kelompok MT menunjukkan erosi dan deskuamasi epitel. Tingkat erosi dan deskuamuasi epitel menurun sebanding dengan peningkatan dosis VCO. Pemberian VCO mampu meningkatkan tinggi vili dan tebal mukosa duodenum mencit yang diberi minyak trans. Dosis VCO 600 mg/KgBB efektif menurunkan tingkat erosi dan deskuamasi epitel mukosa duodenum mencit yang diberi minyak trans. Kata kunci: Duodenum, Minyak trans, Tebal mukosa, Tinggi vili, VCO *ABSTRACT* Excessive consumption of trans oil can interfere with the digestive system so that nutrient absorption is inhibited. VCO contains saturated fatty acids which act to bind free radicals and are good antioxidants. Further research is needed on whether VCO is able to repair the damaged digestive tract due to excessive consumption of trans oil. This study aimed to determine the effect of VCO on height villi and duodenal mucosa in mice induced by high doses of trans oil (MT). A total of 25 male mice with the DDY strain aged 2 months were divided into 5 groups, namely Control (-) NaCl 0.9%; Control (+) MT; K1 (MT+VCO 200 mg/KgBW); K2 (MT+VCO 400 m g/KgBW); K3 (MT+VCO 600 mg/KgBW). After 8 weeks of treatment, the duodenum was taken and histological preparations were made. The height of the villi and thick the mucosa of the duodenum were measured with Image-J software in five visual fields. The data were statistically tested using ANOVA at=0.05 and continued with Duncan's test. The morphology of the mucosa of the duodenum was analyzed descriptively. The results of measuring the height villi of the duodenal mucosa that were given the MT+VCO 200 mg/KgBW were 517,37±21,78 µm, MT+VCO 400 mg/KgBW were 574,9±26,61 µm and MT+VCO 600 mg/KgBW were 651,1±41,74 µm. The thickness of duodenal mucosa that give MT+VCO 200 mg/KgBW were 525,61, MT+VCO 400 mg/KgBW were 556,6±20,22 µm and MT+VCO 600 mg/KgBB were 614,2±29,20 µm.). The height villi of duodenal and thickness of duodenal mucosa significantly than the MT group (sig<0.05). The structure of the duodenal mucosa in the MT group showed erosion and desquamation of the epithelium. The rate of epithelial erosion and desquamation decreased in proportion to the increase in VCO dose. The administration of VCO was able to increase the villi height and thickness of the duodenal mucosa of mice treated with trans oil. The dose of VCO 600mg/kg BW was effective in reducing the erosion and desquamation of the duodenal mucosal epithelium of mice treated with trans oil.
