Abstract

Proses perbanyakan tanaman pisang secara in vitro melibatkan serangkaian tahapan yang penting untuk mencapai produksi sekunder plantlet pisang dalam jumlah yang signifikan dan seragam. Percobaan ini dilaksanakan mulai bulan Desember 2022 – Juni 2023 di Laboratorium Kultur Jaringan, Universitas Negeri Jakarta dengan tujuan untuk mengetahui pengaruh dan konsentrasi optimal pemberian kombinasi ZPT pada induksi dan multiplikasi tunas Pisang Raja Bulu. Tahap awal fokus pada induksi tunas pisang dengan menggunakan enam kombinasi IAA dan BA sebagai faktor penentu. Tahap multiplikasi dilakukan dengan menambahkan dua konsentrasi TDZ sebesar 0,22 mg/L dan 0,44 mg/L pada enam perlakuan IAA dan BA. Hasil percobaan menunjukkan pemberian 0,2 mg/L IAA dan 6,75 mg/L BA dapat menginduksi waktu kemunculan tunas dengan cepat yaitu pada rata-rata 4,40 ± 1,11 hari. Sedangkan pada tahap multiplikasi tunas, pemberian 0,5 mg/L IAA + 2,25 mg/L BA + 0,22 mg/L TDZ dapat memberikan rerata jumlah tunas dan akar terbanyak sebesar 5,60 ± 2,82 tunas dan 8,80 ± 2,04 akar. Selain itu, pemberian 0,5 mg/L IAA + 4,50 mg/L BA + 0,22 mg/L TDZ berpotensi sebagai kombinasi konsentrasi yang baik untuk mendapatkan jumlah daun dan tinggi tanaman dengan rerata sebesar 6,00 ± 0,71 helai dan 7,24 ± 1,34 cm. Hasil statistik menunjukkan bahwa besar pemberian kombinasi IAA, BA dan TDZ berpengaruh nyata namun tidak berbanding lurus terhadap jumlah tunas, jumlah akar, jumlah daun, panjang akar dan tinggi planlet Pisang Raja Bulu yang dihasilkan. *** The process of in vitro propagation of banana plants involves a series of crucial stages to achieve significant and uniform secondary production of banana plantlets. This experiment was conducted from December 2022 to June 2023 at the Tissue Culture Laboratory, Jakarta State University, with the aim of determining the optimal effect and concentration of applying a combination of Plant Growth Regulators (PGRs) on the induction and multiplication of Raja Bulu Banana shoots. The initial stage focused on banana shoot induction using six combinations of IAA and BA as determining factors. The multiplication stage was carried out by adding two concentrations of TDZ at 0.22 mg/L and 0.44 mg/L to the six IAA and BA treatments. The experimental results showed that the application of 0.2 mg/L IAA and 6.75 mg/L BA could induce rapid shoot formation, with an average time of 4.40 ± 1.11 days. Meanwhile, in the shoot multiplication stage, the application of 0.5 mg/L IAA + 2.25 mg/L BA + 0.22 mg/L TDZ yielded the highest average number of shoots and roots, which were 5.60 ± 2.82 shoots and 8.80 ± 2.04 roots. Additionally, the application of 0.5 mg/L IAA + 4.50 mg/L BA + 0.22 mg/L TDZ showed potential as a good combination for obtaining the highest average number of leaves and plant height, which were 6.00 ± 0.71 leaves and 7.24 ± 1.34 cm, respectively. The results of the statistical test indicated that the magnitude of the application of IAA, BA, and TDZ combinations significantly influenced the number of shoots, number of roots, number of leaves, root length, and plantlet height of the produced Raja Bulu banana plantlets.
