Abstract
Sacha inchi (Plukenetia volubilis L.) merupakan tanaman dalam keluarga kacang- kacangan yang tumbuh di wilayah Andes Amerika Selatan. Sacha inchi dikenal dengan potensi antioksidannya yang tinggi, karena mengandung senyawa metabolit sekunder terutama flavonoid. Penelitian ini bertujuan untuk mengetahui kandungan senyawa metabolit sekunder, aktivitas antioksidan terhadap DPPH dan ABTS, dan prediksi gugus fungsi pada ekstrak biji sacha inchi. Pengujian ini menggunakan RAL (Rancangan Acak Lengkap) dengan dua faktor, yaitu variasi ketinggian lahan tanam (100 mdpl, 400 mdpl, dan 1000 mdpl), dan variasi pelarut (ethanol 96%, n-heksana, dan etil asetat). Analisis data aktivitas antioksidan menggunakan ANOVA dua arah dengan uji lanjutan Duncan. Hasil skrining fitokimia menunjukkan bahwa senyawa yang terkandung dalam biji sacha inchi pada setiap variasi ekstrak dari interaksi antara pelarut dan ketinggian lahan tanam yaitu alkaloid, flavonoid, saponin, dan tannin. Hasil pengujian antioksidan terhadap DPPH menunjukkan hasil bahwa ketinggian lahan tanam 400 mdpl yang paling optimal dalam meredam radikal bebas dengan nilai IC50 ≤ 50 ppm,yaitu sebesar 60,91 ppm, pada ketinggian 100 mpdl 27,64 ppm, dan di 1000 mdpl 16,39 ppm. Hasil pengujian antioksidan terhadap ABTS menunjukkan hasil bahwa ketinggian lahan tanam 400 mdpl yang paling optimal dalam meredam radikal bebas IC50 ≤ 50 ppm yaitu sebesar 38,39 ppm, pada ketinggian 100 mpdl 10,59 ppm, dan di 1000 mdpl 15,45 ppm. Gugus fungsi dugaan yang terdapat pada biji sacha inchi dengan tiga variasi ketinggian yaitu O-H, C-H, C=C, C-O, dan N-H yang mengindikasikan adanya senyawa alkaloid, flavonoid, dan tanin. **** Sacha inchi (Plukenetia volubilis L.) is a plant in the legume family that grows in Andes of South America. Sacha inchi is known for its high antioxidant potential, because it contains secondary metabolites, especially flavonoids. This study aims to determine the content of secondary metabolites, antioxidant activity against DPPH and ABTS, and the form of chemical bonds in sacha inchi seed extract. This test used RAL (Completely Randomized Design) with two factors, namely variations in the height of the planting area (100 mdpl, 400 mdpl, and 1000 mdpl), and variations in solvents (96% ethanol, n-hexane, and ethyl acetate). Analysis of antioxidant activity data used two-way ANOVA with Duncan's follow-up test. The results of the phytochemical screening showed that the compounds contained in sacha inchi seeds in each variation of the extract from the interaction between the solvent and the height of the planting area were alkaloids, flavonoids, saponins, and tannins. The results of testing for antioxidants on DPPH showed that the height of the planting area 400 mdpl was the most optimal in reducing free radicals with an IC50 value ≤50 ppm, which was 60.91 ppm, at an altitude of 100 mpdl 27.64 ppm, and at 1000 mdpl 16.39 ppm. The results of antioxidant testing for ABTS showed that the height of the planting area 400 mdpl was the most optimal in reducing free radicals IC50 ≤ 50 ppm, namely 38.39 ppm, at an altitude of 100 mpdl 10.59 ppm, and at 1000 mdpl 15.45 ppm. Presumed functional groups found in sacha inchi seeds with three variations in height is O-H, C-H, C=C, C-O, and N-H which indicated the presence of alkaloids, flavonoids, and tannins.
