Abstract
Paparan radiasi terhadap pekerja radiasi dapat mengakibatkan kerusakan jaringan hingga menyebabkan putusnya double strand pada DNA. Putusnya double strand DNA akibat radiasi akan menyebabkan serin 139 pada histon H2AX mengalami fosforilasi sehinggaterbentuk histon γ-H2AX. Histon ɣ-H2AX merupakan biomarker adanya kerusakan pada double strand DNA. Tujuan penelitian ini adalah untuk medeteksi respon seluler awal pada limfosit darah pekerja radiasi yang diradiasi dengan sinar gamma 2 Gy. Limfosit diisolasi dari darah perifer, yang diradiasi. Pemisahan limfosit dilakukan dengan menambah larutan histopaque. Kerusakan double strand DNA dideteksi dengan reaksi antibodi histon γ-H2AX terhadap protein histon γ-H2AX pada limfost dan pengamatan dilakukan setelah inkubasi selama 24 jam, 48 jam, dan 72 jam. Hasil penelitian menunjukan bahwa jumlah histon ɣ-H2AX pada limfosit paska-irradiasi mengalamai peningkatan dibandingkan jumlah histon ɣ-H2AX pada limfosit pra-radiasi. Inkubasi pada limfosit paska irradiasi selama 24 jam, 48 jam, dan 72 jam, menunjukkan penurunan jumlah histon ɣ-H2AX, namun penurunan signifikan jumlah histon ɣ-H2AX terjadipada inkubasi selama 24 jam. ************************************** Radiation exposure to radiation workers can cause tissue damage to cause a break in the double strand of DNA. The breakage of the double strand DNA due to radiation will cause serine 139 in histone H2AX to be phosphorylated to form histone γ-H2AX. Histone γ-H2AX is a biomarker of damage to the double strand DNA. The purpose of this study was to detect early cellular responses in blood lymphocytes of radiation workers irradiated with 2 Gy gamma rays. Lymphocytes were isolated from irradiated peripheral blood. Lymphocyte separation was done by adding histopaque solution. DNA double strand damage was detected by the reaction of histone γ-H2AX antibodies against histone γ-H2AX proteins in lymphosts and observations were made after incubation for 24 hours, 48 hours, and 72 hours. The results showed that the number of histone -H2AX in post-irradiated lymphocytes increased compared to the number of histone γ-H2AX in pre-irradiated lymphocytes. Incubation of lymphocytes after irradiation for 24 hours, 48 hours, and 72 hours, showed a decrease in the number of histone γ-H2AX, but a significant decrease in the number of histone γ-H2AX that occurred during incubation for 24 hours.
