Abstract

Hiperkolesterolemia merupakan suatu keadaan dimana kosentrasi kolesterol didalam darah melebihi batas normal. Pencegahan hiperkolesterolemia dapat dilakukan dengan modifikasi diet. Susu sinbiotik merupakan produk pangan olahan susu yang difermentasikan oleh bakteri asam laktat (Lactobacillus acidophilus) dengan penambahan prebiotik untuk memberikan efek yang lebih baik. Penelitian ini bertujuan mengetahui pengaruh pemberian susu sinbiotik (SS) terhadap kadar kolesterol pada mencit (Mus musculus) model hiperkolesterolemia menggunakan minyak trans (MT). Metode penelitian yang digunakan adalah eksperimental, dengan desain post test only with control group. Sebanyak 30 ekor mencit jantan strain DDY berumur 2 bulan dibagi menjadi 6 kelompok yaitu kelompok MT (kontrol-); MT+simvastatin (kontrol+); MT+SS 0%; MT+SS 1%; MT+SS 3% dan MT+SS 5%. Data profil lipid yang diukur meliputi kadar kolesterol total, trigliserida, High Density Lipoprotein (HDL) dan Low Density Lipoprotein (LDL) serum darah mencit. Berdasarkan hasil yang didapatkan kolesterol total, LDL mengalami penurunan dan HDL mengalami peningkatan, akan tetapi hasil oneway ANOVA tidak terdapat pengaruh signifikan (P>0,05). Hasil pengukuran kadar trigliserida pada kelompok 1%;3% dan 5% tepung pisang tanduk sebesar 100.1±14.54; 70.8±20.99; dan 47.4±9.57. Berdasarkan hasil uji oneway ANOVA menunjukan bahwa terdapat pengaruh signifikan penambahan tepung pisang tanduk terhadap penurunan kadar trigliserida dengan nilai signifikansi (P<0,05). Uji DMRT menunjukan bahwa susu sinbiotik dengan 5% tepung pisang tanduk dosis 200mg/dL merupakan konsentrasi terbaik dalam menurunkan kadar trigliserida serum darah mencit dengan nilai penurunan sebesar 62,48% dari kontrol (-) dan menurunkan 44,78% dari kontrol (+). *********************************************************** Hypercholesterolemia is a condition in which the concentration of cholesterol in the blood exceeds the normal limit. Prevention of hypercholesterolemia can be done by modifying the diet. Synbiotic milk is a dairy product that is fermented by lactic acid bacteria (Lactobacillus acidophilus) with the addition of prebiotics to provide a better effect.. This study aims to determine the effect of giving synbiotic milk (SS) on cholesterol levels in mice (Mus musculus) hypercholesterolemic model using trans fatty acids (MT). The research method used is experimental, with a completely randomized design. A total of 30 male mice with the DDY strain aged 2 months were divided into 6 groups, namely the MT group (control-); MT+simvastatin (control+); MT+SS 0%; MT+SS 1%; MT+SS 3% and MT+SS 5%. The lipid profile data measured included total cholesterol, triglycerides, High Density Lipoprotein (HDL) and Low Density Lipoprotein (LDL) levels of mouse blood serum. Based on the results obtained, total cholesterol, LDL decreased and HDL increased, but the results of one-way ANOVA had no significant effect (P>0.05). The results of the measurement of triglyceride levels in 1%, 3% and 5% banana flour were 100.1±14.54; 70.8±20.99; and 47.4±9.57. Based on the results of the one-way ANOVA test, it showed that there was a significant effect of the addition of banana flour on the reduction of triglyceride levels with a significant value (P<0.05). DMRT test showed that synbiotic milk with 5% banana flour at a dose of 200mg/dL was the best concentration in reducing blood serum triglyceride levels in mice with a decrease of 62.48% from control (-) and 44.78% from control (+).
