Abstract
Pemuliaan tanaman melalui induksi mutasi merupakan metode efektif dalam meningkatkan keragaman tanaman krisan (Chrysanthemum morifolium) dengan iradiasi gamma secara in vitro. Penelitian ini bertujuan untuk mengetahui pengaruh BAP terhadap multiplikasi tanaman krisan, mengetahui pengaruh ZPT (Zat Pengatur Tumbuh) terhadap induksi kalus krisan mendapatkan lethal dose (LD50) serta mengetahui respon krisan varietas ‘Jayanti Agrihorti’ dan ‘Swarna Kencana’ terhadap dosis iradiasi gamma. Penelitian dilaksanakan menggunakan Rancangan Acak Lengkap (RAL) dengan 3 percobaan: (1) Multiplikasi krisan pada beberapa dosis ZPT (BAP 0 mg/l dan 0,1 mg/l), (2) Induksi kalus krisan dari eksplan daun (NAA 0,5 mg/l + BAP 0,5 mg/l; NAA 0,05 + BAP 2; Pikloram 0,5 mg/l + BAP 0,5 mg/l; 2,4D 2 mg/l + Kinetin 0,5 mg/l; 2,4D 1 mg/l + BAP 1 mg/l; 2,4D 2 mg/l + BAP 2 mg/l), (3) Induksi mutasi eksplan daun krisan dengan iradiasi gamma (0, 10, 15, 20, 25, 30 Gy). Hasil menunjukkan bahwa dosis BAP 0,1 mg/l merupakan konsentrasi optimum untuk menghasilkan jumlah tunas dan daun terbanyak. Dosis ZPT terbaik untuk menginduksi pertumbuhan kalus adalah NAA 0,5 mg/l + BAP 0,5 mg/l dan NAA 0,05 + BAP 2. Nilai LD50 krisan var. Swarna Kencana pada 25,50 Gy, dengan persamaan regresi Y = 8,84 – 3,11x – 5,37x2 yang dianalisis dengan Curve Expert 1.4. Pada krisan varietas Jayanti Agrihorti tidak terdapat pertumbuhan tunas, serta jumlah akar tertinggi pada dosis 0 Gy, jumlah akar lebih rendah seiring bertambahnya dosis iradiasi gamma. Jumlah tunas dari kalus krisan var. Swarna Kencana pada dosis 0 Gy (kontrol) menunjukkan pertumbuhan tertinggi, dosis iradiasi 20-30 Gy menghambat regenerasi tunas krisan.*****Plant breeding through mutation induction is an effective method in increasing the diversity of chrysanthemum (Chrysanthemum morifolium) plants with gamma irradiation in vitro. This study aims to determine the effect of BAP on chrysanthemum plant multiplication, determine the effect of ZPT (Plant Growth Regulator) on chrysanthemum callus induction to obtain a lethal dose (LD50) and determine the response of chrysanthemum varieties 'Jayanti Agrihorti' and 'Swarna Kencana' to gamma irradiation doses. The research was conducted using a Completely Randomized Design (CRD) with 3 experiments: (1) Multiplication of chrysanthemums at several doses of PGR (BAP 0 mg/l and 0.1 mg/l), (2) Induction of chrysanthemum callus from leaf explants (NAA 0.5 mg/l + BAP 0.5 mg/l; NAA 0.05 + BAP 2; Picloram 0.5 mg/l + BAP 0.5 mg/l; 2,4D 2 mg/l + Kinetin 0.5 mg/l; 2,4D 1 mg/l + BAP 1 mg/l; 2,4D 2 mg/l + BAP 2 mg/l), (3) Induction of mutations in chrysanthemum leaf explants with gamma irradiation (0, 10, 15, 20, 25, 30 Gy). The results showed that a dose of BAP 0.1 mg/l was the optimum concentration to produce the highest number of shoots and leaves. The best dose of PGR to induce callus growth is NAA 0.5 mg/l + BAP 0.5 mg/l and NAA 0.05 + BAP 2. The LD50 value of chrysanthemum var. Swarna Kencana at 25.50 Gy, with the regression equation Y = 8.84 – 3.11x – 5.37x2 analyzed by Curve Expert 1.4. In chrysanthemum variety Jayanti Agrihorti there is no shoot growth, and the highest number of roots is at a dose of 0 Gy, the number of roots is lower as the gamma irradiation dose increases. The number of shoots from chrysanthemum callus var. Swarna Kencana at a dose of 0 Gy (control) shows the highest growth, an irradiation dose of 20-30 Gy inhibits chrysanthemum shoot regeneration.
