Abstract

ABSTRAK Flt3 Receptor Interacting Lectin(FRIL) adalah protein lektin spesifik manosa/glukosayang mempunyai kemampuan untuk berikatan dengan antibodi kelas IgM. Antibodi kelas IgM merupakan antibodi yang muncul atau terekspresi ketika suatu antigen pertama kali masuk kedalam tubuh. Antigen adalah benda asing dari luar tubuh yang dapat merangsang respon imun, contohnya adalah bakteri dan virus. Respon imun bersifat spesifik, dimana komplemen hanya akan terbentuk jika antibodi dapat mengikat antigen.Kemampuan FRIL untuk mengikat IgM menunjukan potensi FRIL sebagai marker infeksi suatu virus. Penelitian ini merupakan penelitian pendahuluanyang bertujuan untuk mengisolasi dan mengkarakterisasi protein FRILkacang komak sebagai sampel untuk uji reaksi FRIL dengan IgMyang terbentuk akibat infeksi virus tertentu. Hasil isolasi protein FRILmenggunakan matriks Sephadex G-100 dengan teknik Kromatografi KolomFiltrasi Geldidapatkan satupuncakkromatogram yang sama setelah tiga kali pengulangan. Hasil Native gel elektroforesis FRILmemiliki berat molekul 67 kDa dan dengan SDS-PAGE terlihat mempunyai sub unit dan sub-unit . Kata kunci : FRIL, Kacang Komak, Spektrofotometer ABSTRACT Flt3 Receptor Interacting Lectin (FRIL) is a manose / glucose specific lectin protein that has the ability to bind to IgM class antibodies. IgM class antibodies are antibodies that appear or are expressed when an antigen first enters the body. Antigens areforeign bodies from outside the body that can stimulate an immune response, for example bacteria and viruses. The immune response is specific, where complement will only be formed if antibodies can bind to antigens. The ability of FRIL to bind to IgM shows the potential of FRIL as a marker of virus infection. This research is a preliminary study that aims to isolate and characterize coma bean FRIL protein as a sample for the FRIL reaction with IgM reactions formed by certain viral infectionsin the next study. The results of FRIL protein isolation using Sephadex G-100 matrix with the Gel Filtration Column Chromatography technique obtained one peak of the same chromatogram after three repetitions. Native electrophoresis gel test show that FRIL has a molecular weight of 67 kDa and with SDS-PAGE appears to have subunits and β subunits. Keywords: FRIL, Komak Bean, Spectrophotometer
