Abstract
Low Density Polyethylene (LDPE) merupakan salah satu polimer plastik yang sering digunakan dalam kehidupan sehari-hari. Akumulasi LDPE yang sulit didegradasi di lingkungan dapat menyebabkan pencemaran. Remediasi oleh aktinomisetes dapat dijadikan salah satu upaya untuk mengatasi permasalahan tersebut. Tujuan penelitian ini yaitu mendapatkan dan mengetahui kemampuan isolat aktinomisetes pendegradasi plastik LDPE asal Teluk Jakarta ditinjau dari pertumbuhan aktinomisetes, nilai pH, berat kering plastik serta struktur kimia dan fisik plastik. Uji degradasi plastik LDPE dilakukan dengan metode eksperimental menggunakan rancangan acak lengkap faktorial dengan jenis isolat dan waktu inkubasi sebagai perlakuan. Sebanyak 23 isolat aktinomisetes berhasil diisolasi dari sampel sampah plastik dan sedimen asal Teluk Jakarta. Berdasarkan hasil penapisan, diperoleh 3 isolat yang memiliki indeks zona bening yang tinggi, yaitu ACTTJ22-14, ACTTJ22-06 dan ACTTJ22-23. Isolat ACTTJ22-14 merupakan isolat dengan pola pertumbuhan yang relatif baik selama uji degradasi. Hasil ANOVA dua arah dan DMRT 5% menunjukkan semua perlakuan isolat pada waktu inkubasi ke-40 menunjukkan nilai pH yang berbeda nyata terhadap semua perlakuan isolat pada hari ke-0. Hasil ANOVA dua arah dan DMRT 5% menunjukkan waktu inkubasi pada hari ke-40 memberikan pengaruh nyata terhadap persentase kehilangan berat kering plastik LDPE. Isolat ACTTJ22-14 dan konsorsium menghasilkan persentase kehilangan berat kering yang tidak berbeda nyata dengan kehilangan berat kering sebesar 0,310 ± 0,03%. dan 0,298 ± 0,03%. Analisis dengan FTIR menunjukkan terjadinya perubahan gugus fungsi pada plastik yang mengalami degradasi. Analisis fisik permukaan plastik menggunakan SEM menunjukan terjadinya degradasi berupa lubang. Berdasarkan hasil yang diperoleh, aktinomisetes yang diperoleh dari Teluk Jakarta memiliki kemampuan mendegradasi plastik LDPE. **** Low Density Polyethylene (LDPE) is one of the plastic polymers often used in daily life. The accumulation of LDPE that is difficult to degrade in the environment can cause pollution. Remediation by actinomycetes can be used as an effort to overcome these problems. The purpose of this study was to obtain and determine the ability of actinomycetes isolates to degrade LDPE plastic from Jakarta Bay in terms of actinomycetes growth, pH value, plastic dry weight and chemical and physical structure of plastic. LDPE plastic degradation test was conducted by experimental method using factorial complete randomized design with isolate type and incubation time as treatments. A total of 23 actinomycetes isolates were successfully isolated from plastic waste and sediment samples from Jakarta Bay. Based on the screening results, 3 isolates were obtained that had a high clear zone index, namely ACTTJ22-14, ACTTJ22-06 and ACTTJ22-23. ACTTJ2214 isolate is an isolate with a relatively good growth pattern during the degradation test. The results of two-way ANOVA and DMRT 5% showed that all isolate treatments at the 40th incubation time showed significantly different pH values from all isolate treatments on day 0. The results of two-way ANOVA and DMRT 5% showed that the incubation time on day 40 gave a significant effect on the percentage of dry weight loss of LDPE plastic. ACTTJ22-14 isolate and consortium produced a percentage of dry weight loss that was not significantly different from the dry weight loss of 0,310 ± 0,03%. and 0,298 ± 0,03%. Analysis with FTIR showed changes in functional groups in the degraded plastic. Physical analysis of the plastic surface using SEM showed degradation in the form of holes. Based on the results obtained, actinomycetes obtained from Jakarta Bay have the ability to degrade LDPE plastic.
