Abstract
Penyakit bacterial fruit blotch (BFB) yang disebabkan oleh Acidovorax citrulli merupakan penyakit yang menginfeksi tanaman melon (Cucumis melo L.). Pengendalian penyakit ini dapat dilakukan dengan menggunakan pengendali hayati yang diperoleh dari bakteri endofit. Penelitian ini bertujuan untuk memperoleh bakteri endofit asal tanaman melon dan mengetahui potensinya sebagai agen pengendali hayati penyakit BFB. Metode yang digunakan adalah deskriptif, dan metode eksperimental dengan rancangan acak lengkap. Hasil isolasi bakteri endofit dari akar tanaman melon diperoleh sebanyak 11 bakteri endofit. Hasil skrining antagonistic diperoleh 8 isolat mampu menghambat A. citrulli teridentifikasi sebagai Pseudomonas berdasarkan uji fisiologi dan biokimia. Berdasarkan uji DMRT, isolat EB6 memiliki besaran zona hambat tertinggi (4,41 ± 0,28 mm). Hasil uji hipersensitif pada daun tembakau menunjukkan 5 isolat dari total 8 isolat tidak bersifat patogen, dan tiga isolat dengan zona hambat terbesar (EA1, EB4, dan EB6) dipilih untuk diidentifikasi secara molekuler dan uji efektifitas secara in vivo. Analisis secara molekular sekuen pada 16S rRNA menunjukkan bahwa ketiga isolat teridentifikasi sebagai Pseudomonas lactis. Perendaman biji melon yang telah diinfeksi oleh A. citruli kedalam suspensi bakteri endofit P. lactis menunjukkan bahwa P. lactis EB6 memiliki efektivitas tertinggi (70,38%) dalam mengontrol penyakit BFB dan menurunkan keterjadian penyakit sebesar 59,1%. Bakteri P. lactis juga mampu meningkatkan pertumbuhan tanaman melon yang terinfeksi BFB. Berdasarkan uji DMRT, pemberian P. lactis EB4 menunjukkan peningkatan rata-rata pertumbuhan tertinggi pada parameter tinggi tanaman (41,14 ± 1,35 cm), jumlah daun (6,21 ± 0,35) daun, panjang daun (7,01 ± 0,19 cm), dan lebar daun (7,84 ± 0,17 cm) pada 4 MST dibandingkan dengan isolat lain. Spesies bakteri yang diperoleh pada penelitian ini berpotensi digunakan sebagai agen pengendali hayati penyakit BFB pada tanaman melon. Bacterial fruit blotch (BFB) disease caused by Acidovorax citrulli is a disease that infects melon plants (Cucumis melo L.). This disease can be controlled by using biological controllers obtained from endophytic bacteria. This study aimed to obtain endophytic bacteria from melon plants and determine their potential as biological control agents for BFB disease. The method used is a descriptive and experimental method with a completely randomized design. Isolation of endophytic bacteria from the roots of melon found 11 endophytic bacteria. The results of antagonistic screening showed that eight isolates capable of inhibiting A. citrulli were identified as Pseudomonas based on physiological and biochemical tests. Based on the DMRT test, the EB6 isolate had the highest inhibition zone (4.41 ± 0.28 mm). The results of the hypersensitivity test on tobacco leaves showed that five isolates out of a total of 8 isolates were not pathogenic, and three isolates with the most significant inhibition zones (EA1, EB4, and EB6) were selected for molecular identification and in vivo effectiveness test. Molecular analysis using 16S rRNA sequence showed that the three isolates were identified as Pseudomonas lactis. Soaking melon seeds infected by A. citruli into a suspension of endophytic P. lactis bacteria showed that P. lactis EB6 had the highest effectiveness (70.38%) in controlling BFB disease and reduced the incidence of disease by 59.1%. P. lactis bacteria were also able to increase the growth of melon plants infected with BFB. Based on the DMRT test, the administration of P. lactis EB4 showed the highest average growth increase in plant height (41.14 ± 1.35 cm), the number of leaves (6.21 ± 0.35) leaves, leaf length (7.01 ± 0.19 cm), and leaf width (7.84 ± 0.17 cm) at 4 WAP compared with other isolates. The bacterial species obtained in this study have the potential to be used as biological control agents for BFB disease in melon plants.
