Abstract

Pengobatan dermatofitosis atau kurap pada kucing umumnya menggunakan obat yang masih memiliki kandungan kimia dan alkohol yang berbahaya bagi kucing. Efek samping dapat diminimalisir dengan mengganti bahan kimia dengan senyawa aktif antikapang yang dapat dioptimalkan melalui kultur kapang endofit temu kunci. Penelitian ini dilakukan untuk menguji kemampuan antikapang pada kapang endofit temu kunci (Boesenbergia rotunda) koleksi UNJCC sebagai ekstrak gel antiseptik terhadap kapang dermatofitosis kucing ras. Hasil isolasi dengan metode swab dan agar attached didapatkan 2 kapang penyebab dermatofitosis yang terifentifikasi sebagai Microsporum canis pada 3 jenis kucing ras. Hasil uji antagonisme 3 kapang endofit temu kunci koleksi UNJCC memperlihatkan penghambatan pertumbuhan M. canis oleh UNJCC F6 Pleosporales sp. termasuk kategori tinggi sebesar 59,08%. Senyawa aktif antikapang M. canis yang dihasilkan oleh UNJCC F6 Pleosporales sp. diperoleh dengan fermentasi dan dilakukan uji toksisitas. Berdasarkan hasil uji toksisitas senyawa aktif yang dihasilkan merupakan senyawa tidak toksik (LC50>1000ppm). Senyawa aktif diekstrak dan diidentifikasi melalui GC-CM didapatkan senyawa volatil dengan kelompok ester (2 jenis), asam lemak (3 jenis), alkene (2 jenis), fenol (2 jenis), dan alkaloid (1 jenis) yang memiliki aktivitas antimikroba. Ekstrak senyawa aktif ditambahkan pada gel antiseptik untuk menghambat pertumbuhan M. canis. Hasil uji antagonis gel memperlihatkan perlakuan konsentrasi antiseptik senyawa aktif UNJCC F6 tidak berpengaruh secara nyata terhadap pertumbuhan M. canis tetapi memiliki rentang penghambatan 15,19–24,67% dan menunjukkan abnormalitas pada struktur hifa melalui pengamatan mikroskopik. ***** Treatment of dermatophytosis or ringworm in cats generally uses medicines that contain chemicals and alcohol which are dangerous for cats. Side effects can be minimized by replacing chemicals with active antifungal compounds which can be optimized through the culture of fungal endophytes. This research was conducted to test the antifungal ability of the endophytic fungus of fingerroot (Boesenbergia rotunda) from the UNJCC collection as an antiseptic gel extract against the dermatophytic fungal of breed cats. The isolation results using the swab and agar method obtained 2 fungi identified as Microsporum canis that caused dermatophytosis of 3 types of breed cats. The results of the antagonism test for 3 endophytic fungi from the UNJCC collection showed inhibition of the growth of M. canis by UNJCC F6 Pleosporales sp. classified as a high category with a percentage of 59.08%. The active antifungal compound of M. canis produced by UNJCC F6 Pleosporales sp. was obtained by fermentation and carried out toxicity tests. Based on the results of the toxicity test, the active compound produced is a non-toxic compound (LC50>1000ppm). The active compound was extracted and identified through GC-CM, obtained volatile compounds with ester groups (2 types), fatty acids (3 types), alkenes (2 types), phenols (2 types), and alkaloid (1 type) with antimicrobial activity. The active compound extract was added to the antiseptic gel to inhibit the growth of M. canis. The results of the gel antagonist test showed all antiseptic treatment with concentrations of UNJCC F6’s active compounds didn’t significantly affect the growth of M. canis but had an inhibition percentage range of 15.19–24.67% and showed abnormalities in hyphae structure through microscopic observation.
