Abstract

Pengembangan bakteriofag untuk mengatasi penyakit infeksi akibat bakteri menjadi solusi yang potensial, karena bakteriofag mampu menghambat dan merusak biofilm bakteri, contohnya pada Mycobacterium tuberculosis (Mtb). Penelitian ini bertujuan untuk menguji kemampuan bakteriofag asal air limbah yaitu Mt, Cm, dan Kc dalam menghambat dan merusak biofilm bakteri Mycobacterium smegmatis ATCC 14468. Tahapan penelitian yang dilakukan meliputi pembentukan biofilm; penghambatan biofilm dengan metode Minimum Biofilm Inhibitory Concentration; perusakan biofilm dengan metode Minimum Biofilm Eradication Concentration (MBEC); pemberian bakteriofag, antibiotik, dan kombinasinya dengan metode MBEC dan Total Plate Count; visualisasi kerusakan biofilm menggunakan mikroskop cahaya. M. smegmatis ATCC 14468 dikategorikan sebagai bakteri dengan kekuatan produksi biofilm yang sedang berdasarkan nilai indeks biofilm. Nilai penghambatan biofilm terbesar yaitu 70.04% dihasilkan oleh bakteriofag Cm. Hasil uji analisis variansi (ANAVA) tiga arah menunjukkan kombinasi umur biofilm, isolat bakteriofag, dan pengenceran titer bakteriofag berpengaruh signifikan terhadap perusakan biofilm. Hasil ANAVA dua arah menunjukkan kombinasi bakteriofag dan antibiotik tidak berpengaruh signifikan terhadap perusakan biofilm. Penurunan persentase CFU.mL-1 biofilm terbesar yaitu 80,82% dihasilkan oleh kombinasi bakteriofag Cm dan antibiotik etambutol. Visualisasi biofilm menunjukkan kerusakan biofilm akibat bakteriofag Mt, Cm, dan Kc. Ketiga bakteriofag dapat digunakan sebagai antibiofilm M. smegmatis ATCC 14468 dan dapat dikembangkan untuk penanganan biofilm pada Mtb.*****The development of bacteriophages to treat bacterial infections is a potential solution, as bacteriophage capable of inhibiting and destroying bacterial biofilms, such as Mycobacterium tuberculosis (Mtb). This study aims to test the ability of wastewater-derived bacteriophages, Mt, Cm, and Kc, to inhibit and disrupt bacterial biofilms of Mycobacterium smegmatis ATCC 14468. The research steps included biofilm formation; biofilm inhibition using Minimum Biofilm Inhibitory Concentration method; biofilm destruction using Minimum Biofilm Eradication Concentration (MBEC) method; administration of bacteriophages, antibiotics, and their combinations using MBEC and Total Plate Count methods; visualization of biofilm damage using a light microscope. M. smegmatis ATCC 14468 was categorized as bacterium with moderate biofilm production strength. The highest biofilm inhibition value of 70.04% was produced by bacteriophage Cm. The results of the three-way analysis of variance (ANAVA) showed that the combination of biofilm age, bacteriophage isolate, and bacteriophage titer dilution significantly affected biofilm damage. The results of the two-way ANAVA showed that the combination of bacteriophage and antibiotic did not significantly affect biofilm damage. The greatest reduction in biofilm CFU.mL-1 percentage, 80.82%, was achieved by combination of bacteriophage Cm and antibiotic ethambutol. Biofilm visualization showed damage by bacteriophages Mt, Cm, and Kc. All bacteriophages can be used as antibiofilms for M. smegmatis ATCC 14468 and can be developed for biofilm management in Mtb.
