Abstract

Pengembangan bakteriofag untuk menghambat bakteri Mycobacterium tuberculosis menjadi alternatif yang potensial untuk mengatasi penyakit tuberkulosis. Pengujian secara in vitro pada umumnya menggunakan Mycobacterium smegmatis sebagai model. Penelitian ini bertujuan mengisolasi dan mengkarakterisasi bakteriofag asal air limbah yang mampu melisiskan M. smegmatis ATCC 14468, serta mengetahui aktivitas litik terbaik dari kombinasi jenis bakteriofag dan dosis multiplicity of infection (MOI). Penelitian menggunakan rancangan acak kelompok faktorial 3x3 dengan variabel bebas berupa jenis bakteriofag dan dosis MOI (0,01; 0,1; dan 1). Variabel terikat berupa aktivitas litik (PhageScore) dianalisis melalui ANOVA dua arah signifikansi 95% dan uji lanjut DMRT. Tiga jenis bakteriofag, yaitu Kc, Cm, dan Mt berhasil diisolasi dari air limbah. Ketiga bakteriofag tersebut memiliki karakteristik serupa, yaitu spesifik terhadap M. smegmatis ATCC 14468, tidak mampu menginfeksi sel inang pada fase lag, dan hanya stabil pada pH 7. Tingkat stabilitas bakteriofag Mt dapat mencapai suhu 45°C, sementara pada bakteriofag Kc dan Cm pada suhu 37°C. Aktivitas litik terbaik dimiliki oleh bakteriofag Cm pada dosis multiplicity of infection (MOI) 1 dengan nilai PhageScore sebesar 0,6920. Bakteriofag yang berhasil diisolasi memiliki karakteristik yang rentan terhadap aspek fisiologis manusia, seperti suhu dan pH. Perlu optimasi lebih lanjut terhadap ketiga bakteriofag yang ditemukan sehingga karakteristik dan aktivitasnya menjadi lebih baik. Penelitian ini berhasil mendapatkan bakteriofag asal air limbah yang bersifat litik terhadap M. smegmatis dan bakteriofag Cm berpotensi dikembangkan sebagai agen terapeutik untuk penyakit tuberkulosis.*****The development of bacteriophages to inhibit Mycobacterium tuberculosis is a potential alternative for treating tuberculosis. In vitro testing generally uses Mycobacterium smegmatis as a model. This study aimed to isolate and characterize bacteriophages from wastewater capable of lysing M. smegmatis ATCC 14468, and to determine the optimal lytic activity from the combination of bacteriophage type and Multiplicity of Infection (MOI) doses. The study used a 3x3 factorial randomized block design with the independent variables being the type of bacteriophage and the MOI dose (0.01, 0.1, and 1). The dependent variable, lytic activity (PhageScore), was analyzed using a two-way ANOVA with 95% significance and Duncan's Multiple Range Test (DMRT) for post-hoc analysis. Three types of bacteriophages, namely Kc, Cm, and Mt, were successfully isolated from wastewater. These three bacteriophages shared similar characteristics: they were specific to M. smegmatis ATCC 14468, unable to infect host cells during the non-logarithmic (lag) phase, and were only stable at pH 7. The stability of bacteriophage Mt could reach a temperature of 45°C, while bacteriophages Kc and Cm were stable at 37°C. The best lytic activity was exhibited by bacteriophage Cm at a Multiplicity of Infection (MOI) of 1, with a PhageScore of 0.6920. The isolated bacteriophages have characteristics that are susceptible to human physiological conditions, such as temperature and pH. Further optimization of bacteriophages is necessary to improve their characteristics and activity. This research successfully isolated a wastewater-derived bacteriophage lytic to M. smegmatis with the potential to be developed as a therapeutic agent for tuberculosis, especially bacteriophage Cm.
